Real-Time PCR检测Bcl-2和Bax基因在脊髓钝挫伤大鼠中的表达
The Gene Expression of Bcl-2 and Bax in Spinal Cord Contusion Rat Deteced by Real-time PCR
【摘要】 目的使用Real-Time PCR检测脊髓钝挫伤后不同时间点Bcl-2和Bax基因表达变化。方法 SD成年雌性大鼠72只,随机分为假手术组、脊髓挫伤6h组、脊髓挫伤12h组、脊髓挫伤1d组、脊髓挫伤3d组、脊髓挫伤5d组、脊髓挫伤7d组、脊髓挫伤14d组和脊髓挫伤28d组。大鼠于术后相应各时间点取损伤段脊髓进行RealTime PCR,对基因Bcl-2和Bax表达进行定量分析。结果 Real-Time PCR发现假手术组Bax和Bcl-2基因mRNA水平较高,损伤后均明显下降。Bax基因mRNA在损伤后6h立即下降,12h时明显上升(P<0.01),在损伤后3d又呈现显著性下降(P<0.01),此后一直持续这个水平至28d。Bcl-2基因在损伤后6h也明显下降(P<0.01),12h时立即上升(P<0.01),1d时下降(P<0.01),至损伤后14d又显著性上升(P<0.01)。结论脊髓钝挫伤后大鼠Bax和Bcl-2基因mRNA表达水平于早期显著下降,之后明显上升随后又下降。Bax和Bcl-2基因在脊髓损伤后基因的表达变化为脊髓损伤机制研究提供理论依据。
【Abstract】 Objective This study aimed to investigate Bcl-2and Bax gene expression change at different time points following contusive spinal cord injury.Methods A total of 72 adult female SD rats were randomly divided into nine groups:sham group;spinal cord contusion 6h,12 h,1d,3d,5d,7d,14 dand 28dgroups.Real-time PCR was used to investigate the Bcl-2and Bax gene at 6h,12 h,1d,3d,5d,7d,14 dand 28dafter injury.Results Real-time PCR found that the mRNA levels of Bcl-2and Bax in sham group were high,however,they both decreased immediately after injury.At 6hfollowing injury,the expression of Bax gene was significantly decreased(P<0.01)when compared with sham group.While Bax gene level was then increased at 12 hand the difference between 6hgroup and 12 hgroup was statistically sigificant.Moreover,an increasing expression of Bax was discovered in 3dgroup(P<0.01)and it contained a similar level until 28 dafter injury.On the other hand,Bcl-2gene significanly decreased in spinal cord contusion 6hgroup compared with sham group(P<0.01),and immediately increased at 12 hafter injury(P<0.01),then decresed again at 1d(P<0.01).Untill at 14 dfollowing injury the expression of Bcl-2gene was significantly increased(P<0.01).Conclusion After spinal cord injury,the gene expression of Bcl-2and Bax were indeed changed and these suggested that Bcl-2and Bax gene expression change involved in the mechanism of spinal cord contusion.
