浅谈CADM1在骨肉瘤细胞中的表达及其与MMP-2、MMP-9的关系
CADM1 expression in osteosarcoma cells and its relationship with MMP-2 and MMP-9
【摘要】 目的 :探讨CADM1在骨肉瘤细胞中的表达及其与MMP-2、MMP-9的关系。方法 :用Western Blot法检测h FOB1.19人SV40转染成骨细胞及骨肉瘤细胞Saos-2、U-2os、MG-63中CADM1、MMP-2、MMP-9的表达情况,使用图象分析软件Image—Pro Plus 5.0对以上四种细胞中三种蛋白的表达情况进行半定量测定,并运用统计学方法检测在骨肉瘤细胞中CADM1的表达情况及其与MMP-2、MMP-9的关系。结果 :与h FOB 1.19人SV40转染成骨细胞相比,骨肉瘤细胞Saos-2、U-2os、MG-63中CADM1的表达水平明显降低、MMP-2的表达水平明显升高。骨肉瘤细胞Saos-2、MG-63中MMP-9的表达呈缺失状态。CADM1与MMP-2的表达具有负相关性,与MMP-9的表达不具有相关性。结论 :CADM1在骨肉瘤细胞中的表达呈明显降低的表现,其可能与MMP-2、MMP-9在骨肉瘤的发病机制中发挥着重要的作用。
【Abstract】 AIM :To investigate the expression of CADM1 in osteosarcoma cells, and its relationship with MMP-2 and MMP-9.METHODS: The protein levels of CADM1,MMP-2 and MMP-9 in 1 osteoblast and 3 human osteosarcoma cell lines are detected by Western blotting. The 3 kinds protein levels in 4 cell lines are measured by Image—Pro Plus 5.0. Statistical methods is used to investigate the relationship between CADM1 and MMP-2. RESULTS: In Osteosarcoma cell lines Saos-2,U-2os and MG-63,the protein level of CADM1 is Significantly lower than h FOB 1.19 osteoblasts, and the level of MMP-2 is higher In contrast.the expression of MMP-9 is missing in Saos-2 and MG-63.In addition,the expression of CADM1 and MMP-2 are negative correlation, and the expression of CADM1 and MMP-9 are zero correlation. CONCLUSION: The expression of CADM1 in osteosarcoma cells is significantly reduced. CADM1 may play important roles in the development of osteosarcoma with MMP-2 and MMP-9 together.
