HBV X蛋白对阿霉素诱导的肝癌细胞凋亡的影响研究

【摘要】 目的探讨HBV X蛋白对阿霉素处理后的Hep G2细胞凋亡影响及可能机制。方法用阿霉素处理Hep G2、Hep G2/GFP、Hep G2/GFP-HBx细胞,分别用台盼兰染色和流式细胞仪检测各组细胞凋亡率;并通过RT-PCR和Western blot检测P53、PTEN的基因及蛋白表达。结果阿霉素处理36小时后,Hep G2/GFP-HBx细胞凋亡率明显低于另外两组(P<0.01);Hep G2/GFP-HBx细胞PTEN mRNA和蛋白水平均低于另外两组(P<0.01),而p53 mRNA和蛋白水平与另外两组无明显差别(P>0.05)。结论 HBV X蛋白能够抑制阿霉素诱导的Hep G2细胞凋亡和PTEN的表达,HBV X蛋白可能通过影响p53-PTEN环路而抑制细胞凋亡。

【Abstract】 Objective To investigate the effect of hepatitis B virus X protein in the Adriamycin-induced Apoptosis of Hep G2 Cells and its possible mechanism. Methods Hep G2,Hep G2 / GFP and Hep G2 / GFP-HBx cells were treated with adriamycin,and apoptotic cell death were examined by trypan blue exclusion and flow cytometry analysis. The transcription of p53,PTEN was analyzed by RT-PCR,and the expression of p53,PTEN was analyzed by western blot. Results After treated with adriamycin 36 hours,the apoptosis rate of Hep G2 / GFPHBx was significantly lower than Hep G2 cells and Hep G2 / GFP cells; the transcription and expression of PTEN in Hep G2 / GFP-HBx cells decreased,but there was no significant change in the transcription and expression of p53 in Hep G2 / GFP-HBx cells. Conclusions HBV X protein inhibited apoptosis of Hep G2 cells induced by adriamycin and PTEN expression. The inhibitory effect of HBV X protein on the cell apoptosis may be related to the inhibition of p53-PTEN pathway.
