纳米雄黄抑制肿瘤细胞增殖的体内外研究
Realgar nanoparticles inhibiting the proliferation of tumor cells in vitro and in vivo
【摘要】 目的观察雄黄粒径与抑制肿瘤细胞增殖效应之间的相关性。方法传统方法制备水飞雄黄,气流粉碎机制备气流雄黄,微射流制粒机制备微射流雄黄,对3种雄黄颗粒进行电镜观察、粒度测定和X射线衍射测定;显微镜观察3种粒径雄黄对SMMC7721肝癌细胞生长形态的影响;MTT法观察雄黄对SMMC7721细胞增殖的抑制作用;成功建立HepA小鼠实体瘤模型,以生理盐水灌胃作为对照,观察3种粒径雄黄给HepA小鼠灌胃后移植瘤块的质量和坏死率。结果 3种方法制备的雄黄颗粒粒子直径的d50分别为49.26、4.88和0.28μm,以微射流方法制备的样品粒度最小,达到纳米级别;3种颗粒均可使SMMC7721细胞形态发生凋亡变化;MTT法测得的A值分别为0.402±0.036、0.278±0.024和0.183±0.021,对照组为0.634±0.036,F=430.900,P<0.001;瘤块质量分别为(0.425 8±0.230 8)、(0.348 2±0.153 1)和(0.307 7±0.116 8)g,阴性对照组为(0.634 7±0.208 5)g,F=3.76,P=0.028;瘤块坏死率分别为(25.5±19.8)%、(32.7±12.4)%和(37.2±11.8)%,阴性对照组为(18.0±6.5)%,F=2.94,P=0.041。结论雄黄在体内外均对癌细胞有抑制生长和促进凋亡的作用,且随着粒度的降低,作用逐渐增强。
【Abstract】 OBJECTIVE To investigate the relationship between size particles of realgar and inhibition of tumor cell viabilities.METHODS The realgar particles were prepared with three kinds of methods:water-squelch,airflow,and microjet.Firstly electron microscopy observation,particle size measurement and X-ray diffraction were carried out on the three kinds of realgar particles.The size effects of realgar particles on morphology of SMMC7721 cell growth were observed under inverted phase-contrast microscopy.The inhibiting effects on SMMC7721 cell viability were evaluated by MTT assay.The model of HepA solid tumor in mice was succesfully established,with normal saline as control.The weight and necrosis rates of transplanted tumor masses were observed in mice,after intragastric administration of three sizes of realgar particle,respectively.RESULTS The d50 of three kinds of realgar particles were 49.26,4.88 and 0.28μm,respectively.Among the realgar particles prepared by three methods,the smallest particle size was made by the microjet method,which reached the nano level.All of them could induce apoptosis in SMMC7721 cells,and the A values on MTT were 0.402±0.036,0.278±0.024,0.183±0.021 respectively,as the control group it was 0.634±0.036(F=430.900,P<0.001).Tumor weights(g)were 0.425 8±0.230 8,0.348 2±0.153 1and 0.307 7±0.116 8,while the negative control group was 0.634 7±0.208 5(F=3.76,P=0.028).Necrosis rates of tumor were(25.5±19.8)%,(32.7±12.4)%and(37.2±11.8)%,which were considerably higher than that(18.0±6.5)% of negative control group(F=2.94,P=0.041).CONCLUSIONS The realgar could inhibit cancer cell growth and induce cell apoptosis both in vitro and in vivo.With the decrease of the particle size,the effect gradually increased.
