缺氧对结肠癌HT-29细胞Id1表达的影响
Effect of inhibitor of differentiation-1 on expressions of colon carcinoma HT-29 cells under hypoxia environment in vitro
【摘要】 目的:探讨分化抑制因子-1(Id1)在结肠癌细胞缺氧环境下的表达情况。方法:分别在体外1%O2缺氧条件下培养结肠癌HT-29细胞0h、6h、12h、24h及36h后,利用荧光定量聚合酶链反应(real-timePCR)及Westernblot方法检测各时点Id1的mRNA和蛋白表达情况。结果:缺氧能促进HT-29细胞Id1的mRNA和蛋白表达,缺氧12h后Id1的mRNA和蛋白表达至高峰。结论:在一定时限内,缺氧能促进结肠癌HT-29细胞Id1的mRNA和蛋白表达。
【Abstract】 Objective: To explore the Effect of inhibitor of differentiation-1 on expressions of colon carcinoma HT-29 cells under hypoxia environment in vitro. Method: The expressions of Id1 mRNA and protein were detected by fluorescence quantitative polymerase chain reaction(real-time PC R)and W estern blotassay in H T-29 cells under hypoxia 0h、6h、12h、24h、36h environmentin vitro respectively. Results: Hypoxia can promote the expressions of Id1 on mRNA and protein in colon cancer H T-29 cells. Conclusion: H ypoxia can promote the expressions of Id1 on mRNA and protein in colon cancer HT-29 cells.
