CSE和CBS在Hhcy大鼠阴茎海绵体内的表达及其意义

【摘要】 目的 探讨高同型半胱氮酸血症(Hhcy)勃起功能下降大鼠阴茎海绵体内硫化氢(H2S)信号通路及其作用。方法 将20只2月龄雄性健康wistar大鼠适应性喂养1周后随机分为Hhcy组(H组)与对照组(C组),每组10只。给予H组(生理盐水3mL/次+蛋氨酸1g·kg-1·d-1)和C组(生理盐水3mL/次)早晚各1次灌胃并持续6周。6周后采血检测大鼠的血清睾酮、血浆同型半胱氨酸(Hcy)和H2S的浓度。阴茎海绵体内压/平均颈动脉压(ICPmax/MAP)测定后行阴茎海绵体H2S含量及生成率检测,用Western blot和免疫组化法分析阴茎海绵体中胱硫醚-β-合成酶(CBS)与胱硫醚-γ-裂解酶(CSE)的含量与表达。结果大鼠血清睾酮值C组(463.56±142.19)nmol/L,H组(352.76±120.01)nmol/L,两组比较差异无统计学意义(P>0.05)。H组血浆Hcy浓度较C组明显升高[(27.34±2.49)μmol/Lys(11.41±1.50)μmol/L],差异具统计学意义(P<0.05),而其ICPmax/MAP(0、3、5V)均显著低于C组,差异具统计学意义(P<0.05)。血浆H2S浓度、阴茎海绵体内H2S的含量与生成率比较,H组[(13.36±1.75)μmol/L、(57.54±1.99)nmol/mg prot、(1.38±0.24)nmol/(mg·min)]较C组[(22.44±1.53)μmol/L、(88.63±2.05)nmol/mg prot、(4.53±0.33)nmol/(mg·min)]均显著降低,差异均具统计学意义(P<0.05)。阴茎组织中H组CSE和CBS的蛋白含量较C组明显减少,差异均具统计学意义(P<0.05)。结论 阴茎海绵体内CSE和CBS含量减少和表达减弱,内源性H2S含量降低,这可能是Hhcy大鼠阴茎勃起功能下降的原因之一。

【Abstract】 Objective To study the role of hydrogen sulfide(H2S) signaling pathway in the corpus cavernosum of hyperhomocysteinemia(Hhcy) rats with decreased erectile function.Methods Twenty healthy male wistar rats were fed in the same conditions,and a week later,they were randomly divided into two group as the control group(C group,10rats) and Hhcy group(H group,10 rats).By intragastric administration and twice one day,rats in group H were given with methionine(1g·kg-1·d-1) and physiological saline(3mL per time) while the rats in group C with physiological saline(3mLper time).Six weeks later,the concentration of plasma homocysteine(Hcy) and serum testosterone(T) in the rats were measured,and the maximal intracavemous pressure/mean arterial blood pressure(ICPmax/MAP) was observed,the levels of H2S in the blood plasma and penile tissue and the expressions of CSE and CBS in the corpus cavernosum were detected by immunohistochemistry and Western blot.Results There was no significant difference in the level of serum T between the two groups.The concentration of plasma Hey in group H([27.34±2.49]μmol/L) was remarkably higher than that in group C([11.41±1.50]U mol/L)(P<0.05),whereas the ICPmax/MAP(OV,3V,5V) of group H was all significantly decreased(P<0.05).Compared with those in group C,the levels of plasma H2S([22.44±1.53]μmol/L vs[13.36± 1.75]μmol/L,.P<0.05),endogenous H2S in the corpus cavemosal([88.63±2.05]nmol/mg prot vs[(57.54±1.99)]nmol/mg prot,P<.05),endogenous H2S synthesis([4.53±0.33]nmol/mg per min vs[1.38±0.24]nmol/mg per min,.P<0.05) and the contents of CBS and CSE in group H were decreased remarkablly(P<0.05).Conclusion Low expressions of CBS and CSE in the penile corpus cavernosum may result in reduction of endogenous H2 S,which may be one of the mechanisms of reduced erectile function with Hhcy.
