Raptor调节前列腺癌上皮-间质转化机制研究
Raptor may be involve in regulation of EMT in prostate cancer
【摘要】 目的 探讨Raptor调节前列腺癌上皮-间质转化(EMT)作用及机制研究。方法 构建Raptor-shRNA慢病毒载体并筛选具有稳定转染Raptor-shRNA DU145细胞株,应用transwell观察沉默Raptor后DU145细胞株侵袭力的变化,Western Blot观察沉默Raptor后DU145细胞E-cadherin,β-catenin,N-cadherin和vimentin表达,以及RhoA活性表达。结果 成功构建Raptor-shRNA DU145细胞株;transwell检测稳定转染Raptor-shRNA后DU145细胞侵袭力下降(P<0.01);Western Blot检测显示该细胞株E-cadherin和β-catenin表达下降,N-cadherin和vimentin表达升高,RhoA活性表达下降。结论 Raptor与前列腺癌细胞EMT的发生有关,其作用可能是通过RhoA信号途径。
【Abstract】 Objective To analyze the roles of the Raptor in regulation of EMT in prostate cancer and explore its mechanism.Methods Du145 cells with LV- raptor-shRNA were first established.The invasive abilities of the infected Du145 cells with LV-raptor-shRNA were measured by a transwell assay.The expressions of E-cadherin,β-catenin,N-cadherin,vimentin as well as the RhoA activity were detected by Western Blot in Du145 cells with LV- raptor-shRNA.Results The Du145 cells with LV-raptor-shRNA were successfully constructed,the invasive ability of Du145 cells with LVraptor-shRNA was significantly decreased(P<0.01),the expression levels of E-cadherin,β-catenin and the activity of RhoA in Du145 cells with raptor-shRNA were all lower than those of the control cells.However,the expression levels of N-cadherin and vimentin were higher than those of the control cells.Conclusion Raptor may regulate EMT in prostate cancer through activating RhoA signaling pathway.
