玉米交换单株高效筛选体系的研究
Study on improved system of screening recombinants for fine mapping in maize
【摘要】 本研究用改良碱煮法提取玉米叶片基因组DNA,探索了DNA模板量、引物量、PCR保护剂对PCR反应的影响。结果表明,当DNA模板原液稀释5倍,质量浓度为27.3 ng·μL-1,且反应体系中加入保护剂PVP-40和BSA时可有效提高PCR效率,结合双重PCR的方法实现了快速高效的交换单株筛选。
【Abstract】 In this study,a modified alkaline lysis method was used for the genomic DNA extraction from maize leaf,and then the amount of DNA template was studied and the PCR protectants,PVP-40 and BAS,were used. The results show that it was efficient for PCR reaction when DNA template was27. 3 ng·μL- 1and the protectants were used. In addition,the double PCR with the two sets of primers can be used to screen the recombinants with high efficiency.
