BRD4抑制剂JQ1抑制非小细胞肺癌生长的研究
A preliminary study of BRD4 inhibitor JQ1 effects on non-small cell lung cancer cells
【摘要】 目的:探讨BRD4抑制剂JQ1对非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞株及厄洛替尼耐药细胞株生长的影响并探讨其作用机制。方法:用不同浓度JQ1处理不同类型NSCLC细胞株,72 h后用磺酰罗丹明B(SRB)法检测JQ1对细胞增殖的抑制作用,分别用实时定量聚合酶链反应(q RT-PCR)和Western blot法检测癌基因e IF4E的m RNA及蛋白表达水平。结果:JQ1呈浓度依赖性抑制NSCLC细胞株的生长,包括厄洛替尼敏感和耐药细胞株,下调e IF4E的m RNA及蛋白水平表达。结论:JQ1可抑制不同NSCLC细胞株的生长,其作用机制可能与降低e IF4E表达有关。
【Abstract】 Objective:To investigate the effects and mechanism of BRD4 inhibitor JQ1 on the growth of human non-small cell lung cancer(NSCLC)cells including erlotinib resistant cells. Methods:The inhibitory effects of JQ1 on the growth of human NSCLC were observed at different concentrations for 72 hours by sulforhodamine B(SRB)assay. m RNA and protein levels of e IF4 E were detected by quantitative real-time PCR assay and Western blot,respectively. Results:JQ1 inhibited the growth of NSCLC cells including erlotinib parental and resistant cells in a dose dependent manner. The m RNA and protein expression levels of e IF4 E decreased significantly by JQ1 treatment. Conclusion:JQ1 inhibits the growth of NSCLC cells possibly through downregulating e IF4 E expression.
