碱性成纤维细胞生长因子预处理对大鼠脑缺血/再灌注损伤的神经保护作用
Neuroprotective effect of basic fibroblast growth factor pretreatment on cerebral ischemia-reperfusion injury in rats
【摘要】 目的:探讨碱性成纤维细胞生长因子(bFGF)预处理对脑缺血再灌注损伤(IR)的神经保护作用及其可能的机制。方法:90只雄性SD大鼠随机分成假手术组(A组)、脑缺血/再灌注对照组(B组)及bFGF预处理组(C组)。在制备脑缺血/再灌注模型前向C组大鼠侧脑室注射50μg·ml-1的bFGF 5μl,向A组和B组大鼠侧脑室注射等体积的生理盐水,注射2 h后建立脑缺血/再灌注动物模型。分别于术后1、3、7 d进行改良的神经功能评分(mNSS),通过TTC染色计算梗死体积;术后1 d采用甲苯胺蓝染色和TUNEL染色技术观察缺血/再灌注区域神经细胞受损程度和凋亡情况。结果:与B组相比,C组mNSS降低,梗死体积缩小;细胞损伤程度减轻,TUNEL阳性细胞减少。结论:bFGF预处理对大鼠缺血再灌注损伤有神经保护作用,其机制可能与抑制缺血受损区域的神经细胞凋亡有关。
【Abstract】 Objective: To explore the neuroprotective effect and the underlying mechanism of basic fibroblast growth factor( b FGF) pretreatment on cerebral ischemia-reperfusion injury. Methods: A total of 90 healthy male Sprague-Dawley( SD) rats were randomly assigned into three groups: group A( sham-operated group),group B( ischemia-reperfusion control group),and group C( b FGF pretreatment group). 5 μl of b FGF( 50 mg·ml-1in saline) was administered to the animals of group C by intraventricular injection and an equal amount was administered to the animals of groups A and B by intraventricular injection. 2 hours later,ischemia-reperfusion animal model was constructed. The loss of neurological function was assessed by the modified neurological severity score( m NSS) and the infarct volume was determined by TTC staining 1,3,and 7 days after MCAO. Toluidine blue staining( TBS) was performed for gross histological changes and TUNEL staining was performed for detection of cell apoptosis 1 day after MCAO. Results: In animals of group C,the m NSS score and the infarct volume both dropped;the extent of cell damage was alleviated and the number of TUNEL positive cells dropped compared with group B.Conclusion: b FGF pretreatment has a neuroprotective effect on cerebral ischemia-reperfusion injury in MCAO model of rat. The mechanism of pretreated b FGF in neuroprotection might be related to the inhibition of apoptosis of neurons within the ischemic region.
