B7-H4在人喉癌组织中的表达及意义

【摘要】 目的探讨B7-H4在人喉癌组织中的分布以及该分子在人喉癌中表达的意义。方法免疫组化法检测人喉癌组织B7-H4、细胞增殖及上皮间质转化(epithelial mesenchymal transition,EMT)相关标志物分子的表达;免疫双荧光法检测B7-H4与EMT标志物分子的共表达情况。结果免疫组化结果显示检测62例患者中47例B7-H4阳性,阳性率75.81%;B7-H4表达在人喉癌细胞及喉癌组织浸润性淋巴细胞上,主要为胞浆及胞膜表达;此外,人喉癌组织细胞高表达细胞增殖标志物PCNA及EMT标志物如p-Smad2/3、p-Smad3、Vimentin、Snail;免疫双荧光结果显示p-Smad3、Snail、Vimentin与B7-H4共同表达于人喉癌组织细胞。结论 B7-H4广泛表达于人喉癌细胞及组织浸润性淋巴细胞,并与EMT标志物共表达,可能通过促进EMT进程导致喉癌的侵袭及转移,提示其可作为喉癌治疗的一个新靶点。

【Abstract】 To investigate the expression and significance of B7-H4 in human laryngeal carcinoma,immunohistochemistry and double immunofluorescence were used to observe the expression of B7-H4 and EMT markers on human laryngeal carcinoma. The results showed that B7-H4 was expressed on the surface and in the cytoplasm of the human laryngeal carcinoma cells. Furthermore, B7-H4 also expressed on the lymphocytes infiltrated in the human laryngeal carcinoma tissue. But B7-H4 did not expressed on the endothelial cells. In human laryngeal carcinoma, EMT markers were strongly expressed, such as PCNA, CK-18, p-Smad2/3, p-Smad3,Vimentin, Snail. Token together, B7-H4 is widely expressed in human laryngeal carcinoma cells, including cell surface, cytoplasm, and infiltrating lymphocytes. In addition, B7-H4 is co-expressed with EMT markers on tissues from human laryngeal carcinoma. Our findings suggest that B7-H4 can be developed as a novel therapeutic target for human laryngeal carcinoma.
