三维细胞培养条件下膀胱肿瘤细胞能量代谢的初步探讨

【摘要】 目的研究膀胱肿瘤细胞能量代谢的变化。方法应用微流控芯片三维立体培养技术建立成纤维细胞及膀胱肿瘤细胞共培养体系,通过免疫荧光染色方法观察线粒体代谢情况。结果实验组膀胱肿瘤细胞线粒体代谢较对照组活跃,对照组成纤维细胞线粒体代谢较实验组活跃。结论膀胱肿瘤细胞的能量代谢并不完全是"瓦伯格"效应所认为的"即便在氧供充足的条件下肿瘤仍以糖酵解的方式功能",膀胱肿瘤能量代谢也有高效氧化磷酸化的过程,同时也有肿瘤相关细胞有着密切联系,现在有假说认为肿瘤能量代谢是通过诱导毗邻的间质细胞进行糖酵解,引起再循环营养素生成增多。这些高能的代谢产物通过定向转运转移到毗邻的肿瘤细胞并进入三羧酸循环,致使氧化磷酸化增加和足量的ATP生成,从而构成代谢耦联。我们仍需要大量的实验进一步验证并得出更为正确的结论。

【Abstract】 Objective To investigate the energy metabolism of the bladder tumor cell. Methods Buildding a co- culture system of bladder tumor cells and fibroblasts with microfluidic chip,a three- dimensional culture technique. Then,observing the mitochondrial metabolism by immunofluorescence staining method. Results Compared with the control group,the mitochondrial metabolism of the cancer cells was more active in the experimental group. And the fibroblasts shared the performance of the opposite. Conclusions The energy metabolism of bladder tumor cells does not parallele the ‘Warburg effect’as‘even under the condition of sufficient oxygen,cancer cells are still function in the form of glycolysis’. Bladder cancer cells also have efficient oxidative phos-phorylation process. What’s more,it is also closely related to tumor- associated cells,such as fibroblasts.Nowadays,a new hypothesis considered that cancer cells promote the glycolysis of the adjacent interstitial cells,causing increased formation of recycle nutrients. These high- energy metabolites are directional transit transferred to adjacent tumor cells and enter the Krebs cycle. Finally the oxidative phosphorylation is increased and plenty of ATP is produced. We still need a lot of experiments to proving the value of the hypothesis.
