羊栖菜多糖对高脂培养的胰岛β细胞凋亡的影响
Effect of Sargassum Fusiforme Polysaccharide on Apoptosis of Pancreatic Beta Cells in Saturated Fatty Acids
【摘要】 目的研究羊栖菜多糖(Sargassum fusiforme polysaccharide,SFPS)对在高脂培养的胰岛β细胞凋亡的影响及其分子机制。方法采用MTT法检测SFPS对胰岛β细胞增殖的影响,Hoechst 33258检测胰岛β细胞凋亡情况,反转录RCR法检测bax、caspase-3基因表达量的变化。结果与模型组比较,5%SFPS组和10%SFPS组胰岛βTc3细胞的生长抑制明显减轻(P<0.01),胰岛βTc3细胞的凋亡率明显降低(P<0.01),bax和caspase-3基因表达亦降低(P<0.01)。结论 SFPS能抑制高脂诱导的胰岛β细胞凋亡,其机制可能与调控bax和caspase-3基因表达有关。
【Abstract】 Objective To study effect and molecular mechanism of SFPS on Apoptosis of pancreatic beta cells in saturated fatty acids. Methods We observed the cells proliferation by Tetrazolium(MTT) and cell apoptosis of βTc3 cells by Hoechst 33258. We detected the levels of bax and caspase-3 gene expression in all groups by reverse transcription PCR(RT-PCR). Results With comparison to model group,proliferation inhibition rate of βTc3 cells,the apoptotic rate of βTc3 cells the level of bax and caspase-3 gene expression were significantly declined in 5% SFPS group and 10% SFPS group. Conclusion SFPS could inhibite apoptosis of βTc3cells probably through regulating bax and caspase-3 gene expression.
