酶法辅助乙醇优选牡丹种皮总黄酮

【摘要】 研究适合于以牡丹种皮为原料提取总黄酮的方法,对其提取方法工艺条件进行优化。考察了酶种类、浓度、酶解孵育温度、孵育时间和p H值等因素对提取工艺的影响,并在单因素实验基础上,利用正交实验法进行工艺优化。考察结果表明在果胶酶0.05 mg·m L-1、酶解孵育温度45℃、p H4.5条件下进行酶解孵育180 min后,总黄酮得率最高可达74.839 mg·g-1,相对于乙醇提取法牡丹种皮总黄酮得率显著提高。

【Abstract】 We optimized enzyme assisted ethanol extraction of total flavonoids from peony episperm. We studied the effects of factors including enzymolysis temperature,enzyme content,enzymolysis p H,enzymolysis time on the extraction rates of total flavonoids by single-factor and orthogonal test. The optimal extraction conditions were after enzyming pretreatment under the pectinase content of 0. 05 mg·m L- 1,enzymolysis temperature of 45℃,enzymolysis p H of 4. 5,enzymolysis time of 180 min,total flavonoids yield was up to 74. 839 mg · g- 1.Compared with the traditional ethanol extraction,the total flavonoids yield was improved significantly.
