热休克蛋白47和基质金属蛋白酶2在增生性瘢痕皮肤和胎儿皮肤中的表达

【摘要】 目的研究热休克蛋白47(HSP47)和基质金属蛋白酶2(MMP-2)在增生性瘢痕皮肤和胎儿皮肤中的表达及其生物学意义。方法收集妊娠终止的胎儿背部正常皮肤30例,胎龄13~33周;增生性瘢痕组织18例,平均年龄22岁。所有标本用常规石蜡包埋、切片,经HE染色、Masson三色染色和PASM染色,观察组织学结构和胶原纤维的表达;免疫组织化学染色、免疫荧光及免疫印迹试验检测HSP47和MMP-2蛋白在皮肤组织内的表达和定位,RT-PCR检测HSP47和MMP-2 m RNA的水平。结果胎儿皮肤组织内HSP47的表达水平显著低于增生性瘢痕皮肤(P<0.01),而MMP-2表达水平则显著升高(P<0.05)。结论 HSP47和MMP-2对皮肤创面的无瘢痕修复可能具有显著的调节作用。

【Abstract】 Objective To study the expressions of heat shock protein 47(HSP47) and matrix metalloproteinase2(MMP-2) in proliferative scar skin and fetal skin and their potential biological significances. Methods Thirty cases of normal fetal skin in the back of terminated pregnancies with embryonic gestational age of 13 to 33 weeks and eighteen cases of hypertrophic scar tissues from adults with average age of 22 year old were collected. All specimens were embedded in paraffin, sectioned, stained by HE, Masson trichrome and PASM, and the histological structure and the expression of collagen fibers were observed. Immunohistochemistry, immunofluorescence and western blot were used to detect the expressions and localization of HSP47 and MMP-2 protein, RT-PCR was used to detect the m RNA levels of HSP47 and MMP-2. Results The expression of HSP47 in fetal skin was significantly lower than that in hypertrophic scar skin(P < 0.01), but the expression level of MMP-2 was significantly increased(P < 0.05). Conclusion HSP47 and MMP-2 may play an important role in the regulation of skin scarless wound healing.
