桂枝茯苓丸对子宫内膜异位症模型大鼠血管内皮生长因子表达的影响

【摘要】 目的采用皮下移植自体子宫内膜组织的方法建立大鼠子宫内膜异位症模型,观察桂枝茯苓丸对大鼠子宫内膜异位症模型血管内皮生长因子表达的影响。方法 50只大鼠随机分为空白对照、模型组、米非司酮组、桂枝茯苓丸A、B组,其中模型组、米非司酮组及桂枝茯苓丸A、B组均制作大鼠子宫内膜异位症模型。术后桂枝茯苓丸A、B组用桂枝茯苓丸灌胃治疗21 d,米非司酮组灌阳性药米非司酮,空白对照组及模型组灌等体积生理盐水。采用酶联免疫吸附测定(ELISA)法测定大鼠子宫内膜组织VEGF及mRNA、低氧诱导因子-1α(HIF-1α)及HIF-1 mRNA表达,测定血清雌二醇(E2)及白介素-8(IL-8)水平,免疫组织化学法检测细胞增殖核抗原(PCNA)、雌激素受体(ERα)。结果桂枝茯苓丸A、B组VEGF及mRNA、HIF-1α及HIF-1 mRNA在子宫内膜组织上表达下调,E2、IL-8水平及PCNA、ERα表达量均明显降低,与模型组比较,差异均有统计学意义(P<0.05)。结论桂枝茯苓丸可抑制模型大鼠异位内膜的生长与血管生成,减轻子宫内膜异位症血管内皮炎症反应,对实验性大鼠子宫内膜异位症有治疗作用。

【Abstract】 Objective To establish the rat endometriosis model by using the method of subcutaneous transplantation of autologous endometrial tissue,to observe the effect of guizhi fuling pill on the expression of vascular endothelial grow th factors in model endometriosis rats. Methods 50 rats were randomly divided into control group,model group,mifepristone group,guizhi fuling pill A,B group. Model group,mifepristone group and guizhi fuling A,B group were made as rat endometriosis model. Guizhi fuling pill A,B group were given guizhi fuling bolus gavage for 21 d,mifepristone group was given positive medicine danazol,control group and model group were administrated with normal saline. The enzyme linked immunosorbent assay( ELISA) method was used for the determination of rat uterine endometrial tissue VEGF and mRNA,hypoxia inducible factor-1α( HIF-1α) and the expression of HIF-1 mRNA,the determination of serum estradiol( E2) and interleukin-8( IL-8) level,the proliferating cell nuclear antigen( PCNA),estrogen receptor( ERα) were detected by immunohistochemistry. Results The expression of VEGF and mRNA,HIF-1α and HIF-1 mRNA in endometrial tissue in guizhi fuling pill A,B group were dow n regulated,and the expression of E2,IL-8 level and PCNA,ERα volume significant reduced compared with those of model group( P < 0. 05). Conclusion The grow th and angiogenesis of ectopic endometrium guizhi fuling pill can inhibit the grow th of rat endometriosis,reduce vascular endothelial inflammation and has therapeutic effects on experimental rats with endometriosis.
