骨髓间充质干细胞缺血培养上清细胞因子分析
The secreted cytokine concentration changes of bone marrow mesenchymal stem cells(BMSCs)in ischemic conditions
【摘要】 目的 研究骨髓间充质干细胞在缺血条件下分泌的细胞因子浓度变化。方法 用全骨髓差异贴壁法分离培养骨髓间充质干细胞(BMSCs),取第3代细胞进行实验。实验分两组:A组提取正常培养细胞的上清液;B组将细胞置于无血清的培养基中24h后,提取上清液。采用蛋白分析技术检测细胞培养上清液中27种细胞因子变化情况。结果 无血清细胞培养组中ICAM-1、IFN-γ和IL-10的浓度与有血清的细胞培养组比较差异有统计学意义(P<0.05)。结论 骨髓间充质干细胞在缺血条件下分泌的炎症相关细胞因子发生变化。
【Abstract】 Objective To study the secreted cytokine concentration changes of bone marrow mesenchymal stem cells(BMSCs) in ischemic conditions.Methods Bone marrow mesenchymal stem cells were cultured by adherent culture of whole bone marrow.The third generation of cells were used in this study.The experiments were divided into two groups:A.The supernatant wwas collected from normal cell culture.B.The supernatant was from cells in serum-free medium 24 h.27 kinds of cytokines in the supernatant were analyzed through the protein assay.Results Compared with the normal culture condition group,the concentration of ICAM-1,IFN-γ and IL-10 in the serum-free culture group were significant decreased.(P<0.05).Conclusion There were changes in the inflammation related cytokines secreted by bone marrow mesenchymal stem cells in the ischemic condition.
