氯化汞对睾丸间质细胞类固醇激素合成影响的体外研究

【摘要】 [目的]探讨氯化汞(Hg Cl2)对SD大鼠体外培养睾丸间质细胞(leydig cell)类固醇激素合成的影响。[方法]不同浓度Hg Cl2(1×10-8、1×10-7、1×10-6、1×10-5 mol/L)作用于leydig细胞后,测定孕酮含量,运用实时荧光定量逆转录聚合酶链反应(RT-PCR)技术检测Hg Cl2对leydig细胞中St AR、P450scc和3β-HSD m RNA表达的影响。[结果]经不同浓度Hg Cl2染毒后,leydig细胞分泌孕酮的量均有所降低,其中1×10-7、1×10-6、1×10-5 mol/L Hg Cl2染毒组与对照组相比,差异均有统计学意义(P<0.05)。1×10-7、1×10-6和1×10-5 mol/L Hg Cl2剂量组的St AR、P450scc和3β-HSD m RNA相对表达量均低于对照组(P<0.05)。[结论]汞可以影响睾丸间质细胞类固醇激素的合成。

【Abstract】 [Objective] To assess the effects of mercury chloride(Hg Cl2) on steroid hormone biosynthesis in leydig cells isolated from SD rat testis. [Methods] Leydig cells were exposed to various concentrations of Hg Cl2(1×10-8, 1×10-7, 1×10-6, and 1×10-5 mol/L). Progesterone in the supernatant was measured. The levels of m RNA expression of St AR, P450 scc, and 3β-HSD in leydig cells were determined by real-time fluorescence quantitative reverse transcription polymerase chain reaction(RT-PCR). [Results] The progesterone content secreted by Leydis cells was reduced after treated by various concentrations of Hg Cl2, and the progesterone contents in the 1×10-7, 1×10-6, and 1×10-5 mol/L Hg Cl2 groups were significantly lower than that of the control group(P < 0.05). The m RNA relative expression levels of St AR, P450 scc, and 3β-HSD in the leydig cells exposed to Hg Cl2 at the concentration of 1×10-7, 1×10-6, and 1×10-5 mol/L were also statistically lower than that of the control group(P < 0.05). [Conclusion] Mercury could affect steroid hormone biosynthesis of leydig cells.
