麻黄附子细辛汤联合抗生素的体外抗大肠埃希菌作用研究

【摘要】 目的:研究麻黄附子细辛汤与抗生素联合应用对抗生素的增效作用,为研究中药对抗生素增效的作用机制和筛选活性成分奠定实验基础。方法:将各组培养基分别接种大肠埃希菌后37℃培养过夜,空白组不接种细菌,每12小时取出计数,计算活菌数;同时使用连续稀释法检测其各自最小抑菌浓度(MIC)及最低杀菌浓度(MBC);并且用纸片法检测抑菌圈。结果:(1)中药复方组12 h细菌逐渐生长,培养120 h细菌浓度逐渐下降,培养180 h细菌浓度保持不变;头孢唑林组培养12 h细菌数逐渐减少,60 h时细菌减少的趋势停止;中药复方联合头孢唑林组培养12 h细菌数逐渐减少,144 h各组细菌无生长迹象;替加环素药物干预后细菌数急剧下降,但是直到观察结束仍有部分细菌存活,阴性组细菌随着培养时间的逐渐延长呈现缓慢生长趋势,空白组无细菌生长。(2)麻黄附子细辛汤对大肠埃希菌MIC为0.625 mg.m L-1,MBC为2.5mg.m L-1;头孢唑林对大肠埃希菌MIC为0.312 mg.m L-1,MBC为1.25 mg.m L-1;中药复方联合头孢唑林对大肠埃希菌MIC为0.156 mg.m L-1,MBC为2.5 mg.m L-1;替加环素对大肠埃希菌MIC为0.078 mg.m L-1,MBC为0.625 mg.m L-1。(3)中药复方联合头孢唑林抑菌环直径和阳性对照组相近,其次是头孢唑林组,中药单独使用的抑菌环直径小于头孢唑林组.麻黄附子细辛汤与头孢唑林联合用药对大肠埃希菌的抗菌效应均增强,麻黄附子细辛汤与头孢唑林联合用药的抗茵作用表现为协同作用(FIC=0.59)。结论:麻黄附子细辛汤联合抗生素具有更明显的抗菌效应,可能与它激活耐药细菌生长有关。

【Abstract】 Objective: The effect of Mahuang Fuzi Xixin decoction combined with antibiotic on escherichia coli was investigated to study the synergistic mechanism of traditional Chinese medicine on antibiotics and lay the experimental foundation for screening of active ingredients. Method: Each group was cultured overnight at 37 ℃ after inoculation of e. coli respectively. The blank group didn’t inoculate with any bacterium, and then was took out every 12 hours for viable counts. At the same time, the minimal inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were detected by continuous dilution method. And inhibition zone was detected by paper strip method. Result:(1) It was found that bacteria grew gradually in the traditional Chinese medicine compound group at 12 h. The bacteria concentration gradually decreased from 120 h to 180 h. And the bacteria concentration remained the same since 180 h. Bacteria reduced gradually in cefazolin group when cultured for 12 h, and the trend of bacteria reduction ended at 60 h. The bacteria count of traditional Chinese medicine combined with cefazolin group reduced gradually at 12 h. After cultured for 144 h, bacteria of all groups showed no sign of growth. Bacterial count decreased sharply after Tigecycline intervention, but some bacteria survived until the end of the observation, The bacteria of negative group showed a trend of slow growth along with culture time. The blank group didn’t have bacterium growth.(2) The MIC and MBC of Mahuang Fuzi Xixin decoction on e. coli was 0.625 mg.m L-1 and 2.5 mg.m L-1 respectively. The MIC and MBC of Cefazolin on e. coli was 0.312 mg.m L-1 and 1.25 mg.m L-1 respectively. The MIC and MBC of Traditional Chinese medicine combined with cefazolin on e. coli was 0.156 mg.m L-1 and 2.5 mg.m L-1 respectively. The MIC and MBC of Tigecycline on e. coli was 0.078 mg.m L-1 and 0.625 mg.m L-1 respectively.(3) Bacteriostasis Circle diameter in Traditional Chinese medicine combined with cefazolin group was similar as positive control group. Bacteriostasis circle diameter was second in cefazolin group. Traditional Chinese medicine(TCM) used alone had smaller bacteriostasis circle diameter than cefazolin group. MAhuang Fuzi Xixin decoction with cefazolin had an enhanced antibacterial effect on e. coli which showed a synergistic antibiosis effect of MAhuang Fuzi Xixin decoction and antibiotics combination showed the synergy effect(FIC = 0.59). Conclusion: MAhuang Fuzi Xixin decoction combined with antibiotics have more obvious antibacterial effect, which may be relatee to the activation the growth of resistant bacteria.
