乳腺癌组织中TET2突变与5hmC表达的关系

【摘要】 目的:探讨乳腺癌组织中TET2(ten-eleven translocation 2)基因的突变与5-羟甲基胞嘧啶(5hmC)表达的关系。方法:收集100例乳腺癌患者的癌组织标本。用Sanger方法检测乳腺癌组织中的TET2突变情况;用免疫组化方法检测乳腺癌组织中5hmC的表达情况;分析TET2突变与5hmC表达的关系。结果:100例乳腺癌组织中有13种TET2突变,在4例患者的肿瘤组织中发现3种以前未报道过的TET2突变(Ala341Val、Leu1622Phe、Pro1578Tyr),在2例患者的肿瘤组织中均检测到Pro1578Tyr突变。免疫组化结果显示,有TET2突变的乳腺癌组织中的5hmC表达明显低于无TET2突变的乳腺癌组织。结论:乳腺癌中TET2突变导致5hmC水平下降。

【Abstract】 Objective:To explore the relationship between ten-eleven translocation 2(TET2)Mutations and Expression of5 hmC in breast cancer.Methods:TET2 mutations in breast cancer tissues from100 patients were detected by Sanger method.The expression of 5hmC in breast cancer tissues was detected by immunohistochemistry.The relationship between TET2 mutations and expression of 5hmC was analyzed.Results:A total of 13 kinds of TET2 mutations were identified in 100 patients.Among them,3 new types of mutations were identified in 4 patients.They were Ala341 Val,Leu1622Phe and Pro1578 Tyr.Pro1578Tyr was identified in two patients.And immunohistochemical analysis showed that there was a sharp reduction of 5hmC in breast cancer tissues with TET2 mutations than in breast cancer tissues without TET2 mutations.Conclusions:TET2 mutations can lead to decrease of 5hmC level in breast cancer.
