正交设计优选枳椇子中总黄酮的提取工艺

【摘要】 目的研究枳椇子总黄酮的超声提取最佳工艺条件;方法利用超声提取法,以药材目数、料液比、超声温度、超声功率为考察因素,枳椇子中芦丁的含量为考察指标,紫外分光光度法测定(417nm)结果最佳提取工艺:干燥药材(20~40目)中加入14倍量的体积分数70%乙醇溶液,超声功率:160 W、温度:70℃、时间:60min时含量最高,为1.17mg/g。回归方程为A=0.033 3C-0.022 3(r=0.999 3),芦丁在5.84~23.36μg/mL范围内线性关系良好,平均回收率为96.92%。结论超声法提取枳椇子中的芦丁优势明显,有效成分溶出快、用时短、效率高,紫外测定其含量灵明度较高、准确。

【Abstract】 Objective to optimize the extracting method of total flavonoids from Turnjujube Methods extracting total flavonoids from Turnjujube by ultrasonication extraction,considering the size of raw material,material-toliquid ratio,extraction temperature,extraction frequency as affecting factor by L9(34)orthogonal design;using rutimun as a reference substance,the rutimun in Turnjujube was determined by UV absorption wave at 417 nm.Results the best extraction condition by ultrasonication extraction was to use 70%alcohol at 70℃for 1hours with160 W,material-to-liquid ratio 20~40mesh granule,and the resultant total flavonoids yield was 1.17mg/g,equation of regression:A =0.033 3C-0.022 3(r =0.999 3),the linearity of rutimun was obtained with in the range of5.84~23.36μg/mL,the average recovery was 96.92%.Conclusion the method is convenient and reliable,this result provide a suitable method research.
