洛铂对人肺腺癌A549、A549/DDP细胞株抑制作用的研究

【摘要】 目的观察洛铂对体外培养人肺腺癌A549细胞株和耐顺铂的A549/DDP细胞株作用,初步探讨洛铂的抗肿瘤作用机制。方法采用caspase酶抑制剂、MTT法等研究不同浓度、时间的洛铂对A549细胞株和A549/DDP细胞株的作用,分析洛铂的作用机制。结果随洛铂浓度升高、作用时间延长,细胞毒作用增强;耐顺铂细胞株存在同样的规律;加入caspase-3、8抑制剂组存活率高。结论洛铂具有剂量-时间依赖性,可作为耐顺铂肿瘤的备选药物;洛铂诱导A549和A549/DDP细胞凋亡,可能涉及caspase-3、8的活化。

【Abstract】 Objective To observe and compare the function of Lobaplatin on lung cancer cells HTB-56 and HTB-56/DDP and make a preliminary study of Lobaplatin mechanisms. Methods Incubate anti-cisplatin pulmonary cells A549、A549/DDP to a certain time then used MTT assay to measure the survival rate of A549、A549/DDP cells and compare them. Results As lobaplatin dose increased, human lung adenocarcinoma cell line A549, A549/DDP survival rates were significantly lower, but with time-dependent; with the same dosage and timing of lobaplatin,comparison of survival rates were not significantly different between the two kinds of cells. The survival ratios of Caspase-3、8 specific inhibitor groups were higher than the Lobaplatin single group. Conclusion Lobaplatin has a cytotoxic effect on cells of A549 and A549/DDP, and has a dose-time dependence and can be used as alternative drug against cisplatinresistant cancer. Lobaplatin on lung cancer A549 and A549/DDP cells may induce apoptosis and this process may involve the activating of caspase-3,8.
