虎耳草素对长春新碱诱导的神经病理性疼痛的影响

【摘要】 目的:探讨虎耳草素对长春新碱诱导的神经病理性疼痛的影响及其作用机制。方法:利用长春新碱致神经病理性疼痛模型小鼠,腹腔注射虎耳草素100 mg/kg,通过行为学实验测试机械性缩足反射阈值;生化检测氧化应激相关指标丙二醛(MDA)和谷胱甘肽(GSH)的表达;ELISA检测炎症因子肿瘤坏死因子α(TNF-α)和白细胞介素(IL)-1β的表达;原子吸收光谱检测钙离子浓度的变化。结果:虎耳草素能够显著地抑制长春新碱诱导的神经病理性疼痛(CNP)模型小鼠机械性痛觉过敏,且能改善CNP小鼠高氧化应激状态和降低CNP小鼠TNF-α、IL-1β的表达水平及钙离子浓度。结论:虎耳草素拮抗CNP,且该作用机制可能与其抑制氧化应激、降低炎症因子TNF-α、IL-1α的表达及钙离子浓度有关。

【Abstract】 Objective:To investigate the antinociceptive effect of bergenin on vincristine-induced neuropathic pain(CNP) and to explore its mechanisms.Methods:Mechanical withdrawal threshold was tested before and after intraperitoneal administration of 100 mg/kg bergenin in the model mice with vincristine-induced neuropathic pain via behavioral experiment.Expression of oxidative stress markers including malondialdehyde and glutathione was determined biochemically.Ca2+ level was detected by atomic absorption spectrometry.Sciatic nerve production of TNF-α and IL-lβ was quantified by ELISA kits.Results:Bergenin significantly attenuated vincristine-induced mechanical allodynia.Bergenin also ameliorated vincristineinduced high oxidative stress,inhibited the expression of TNF-a and IL-1β,and reduced the Ca2+ level in CNP mice.Conclusion:Bergenin may attenuate vincristine-induced neuropathic pain.The effect might partly attribute to the inhibition of oxidative stress and the decrease in the inflammatory cytokines expression as well as Ca2+ level.
