花荵组织培养与快速繁殖技术研究

【摘要】 以花荵茎基部新抽出的侧芽为外植体,研究影响组培快繁的因素,结果表明:最适启动培养基为MS+6-BA1.0 mg·L-1+NAA0.05 mg·L-1;最适增殖培养基为MS+6-BA 2.0 mg·L-1+NAA0.15 mg·L-1,平均增殖系数3.6;最适生根培养基为1/2MS+NAA0.1 mg·L-1,生根率97.78%;最佳移栽基质为河沙,移栽成活率为96.67%。

【Abstract】 The factors which affected tissue culture and rapid propagation was researched by choosing the lateral bud from basal part of stem of Polemonium chinensis. The result showed that the optimum initial medium was MS + 6- BA1. 0 mg·L- 1+ NAA0. 05 mg· L- 1. The optimum multiplying culture was MS + 6- BA 2. 0 mg·L- 1+ NAA0. 15 mg·L- 1. The average proliferation coefficient was 3. 6. The optimum rooting medium was 1 /2MS + NAA0. 1 mg·L- 1. The rooting percentage was 97. 78%. The best transplanting substrate was river sand. The survival rate of transplanting was 96. 67%.
