不同浓度的碘化丙啶染色对细胞周期分布的影响

【摘要】 研究不同浓度的碘化丙啶(PI)染色对细胞周期分布的影响。293T细胞经0、2、4、6Gy的X射线照射后,用浓度分别为5、10、20、50μg/ml的PI染色后,流式细胞仪检测细胞周期的分布情况。0、2、4、6Gy时,不同浓度PI染色时G1/G0期的比例变化如下:(35.90±0.17)至(37.65±0.42),(18.05±0.40)至(19.08±2.01),(4.24±1.31)至(5.89±1.08),(2.88±0.16)至(4.04±1.06),G2/M期的比例变化如下:(13.36±0.36)至(15.39±0.33),(54.56±0.34)至(58.98±1.46),(83.92±4.50)至(84.80±1.43),(92.29±1.56)至(92.89±1.49);S期的比例变化如下:(45.63±0.97)至(49.74±0.16),(22.45±0.35)至(27.04±0.36),(10.05±0.79)至(11.29±0.11),(3.71±1.06)至(4.99±1.01)。在同一照射剂量下,不管是G1/G0期、S期还是G2/M期,不同浓度PI染色对细胞周期分布没有明显差异(P>0.05);但不管5、10、20μg/ml还是50μg/ml的PI染色,随着照射剂量的增加,G2/M期细胞比例明显增加(P<0.01)。随着照射剂量的增加,G2/M期细胞比例明显增加;但对于同一照射剂量,不同浓度PI染色对细胞周期分布没有明显差异,做周期分布实验时选择5μg/ml的PI染色即可。

【Abstract】 Study on the effect of different concentrations of propidium iodid in cell cycle distribution experiment.After 0,2,4,6Gy irradiation with X- ray and treated with propidium iodideof 5,10,20,50μg / ml,the cell cycle distribution of 293 T cells was evaluatedby the flow cytometer. At 0,2,4,6Gy,after dyed with propidium iodideof different concentrations,the percentage changes of G1 / G0 phase as follows:( 35. 90 ± 0. 17) 37. 65 ± 0. 42,( 18. 05 ±0. 40) 19. 08 ± 2. 01,( 4. 24 ± 1. 31) 5. 89 ± 1. 08,( 2. 88 ± 0. 16) 4. 04 ± 1. 06; the percentage changes of G2 / M phase were:( 13. 36 ± 0. 36) 15. 39 ± 0. 33,( 54. 56 ± 0. 34) 58. 98 ± 1. 46,( 83. 92 ± 4. 50) 84. 80 ± 1. 43,( 92. 29 ± 1. 56) 92. 89 ± 1. 49; the percentage changes of S phase as follows:( 45. 63 ±0. 97) 49. 74 ± 0. 16,( 22. 45 ± 0. 35) 27. 04 ± 0. 36,( 10. 05 ± 0. 79) 11. 29 ± 0. 11,( 3. 71 ± 1. 06) 4. 99 ± 1. 01. At the same dose,when stained with different concentration of PI,both G1 / G0,S phase and G2 /M phase,the percentages were no significant difference( P > 0. 05); But regardless of 5,10,20μg / ml or 50μg / ml of PI,the cells proportion of G2 / M phase increased significantly with the increase of irradiation dose( P < 0. 01). The G2 / M phase cells proportion increased significantly with the increase of irradiation dose; But for the same dose,there was no significantdifference on the cell cycle distribution of different concentration of PI staining,So 5μg / ml of PI is appropriate for cell cycle distribution experiment.
