金鱼Dishevelled3基因的克隆与分化表达研究
A Study on Cloning and Differential Expression of Gene Dishevelled 3 in Goldfish
【摘要】 目的:本文主要以金鱼中的dvl-3基因为研究对象,研究dvl-3在金鱼不同的组织和胚胎发育时期中可能行使的功能。方法:采用RACE克隆技术获得金鱼dvl-3基因的c DNA全长,再利用半定量RT-PCR技术和蛋白免疫印记(Western blot)技术分析研究dvl-3基因在mRNA水平和蛋白水平上的表达差异。结果:在金鱼不同组织与胚胎发育时期,dvl-3基因在mRNA水平和蛋白水平上具有显著的表达差异性。结论:由此可见散乱蛋白dvl-3在金鱼不同的组织和胚胎发育时期中可能行使不同的功能,但其具体的功能及机制还有待进一步研究。
【Abstract】 Objective: Here,our study focused on gene dvl-3 in Carassius auratus. Research the function of dvl-3 proteins during embryonic development stages in Carassius auratus as well as in different tissues. Method: Firstly,we used RACE( rapid-amplification of c DNA ends) clone technology to obtain full-length c DNA of gene dvl-3,and then semiquantitative RT-PCR and western blot were used to analyze the differential expression of dvl-3 in mRNA level and protein level respectively. Results: The results showed that: Semi-quantitative RT-PCR and semi-quantitative western blot results are different in different embryonic development and different tissues. Conclusions: Thus it showed that the function of dvl-3 proteins varied during embryonic development stages in Carassius auratus as well as in different tissues.However,the exact mechanism remained to be further explored.
