脂质生物合成的转录调控、相关靶标确证及新药发现
Transcriptional regulation of lipid biosynthesis, related target validation and drug discovery
【摘要】 脂质生物合成失调是代谢异常的重要方面,与高脂血症、胰岛素抵抗、动脉粥样硬化、骨质疏松及肿瘤等多种疾病密切相关.在转录调控层面上,固醇调节元件结合蛋白和肝脏X型受体二者分别负责脂质生物合成及转运代谢相关蛋白的调控.两条调控信号网络之间既相互关联又互相制约,共同维持体内脂质代谢的平衡.转录共激活因子PGC-1?同时调控SREBP、LXR和ERR?三种转录因子的活性,在脂代谢转录调节方面处于关键枢纽地位.
【Abstract】 Disorders in lipid biosynthesis is a key aspect of metabolic disorders, and is associated with many disease, including hyperlipidemia, insulin resistance, atherosclerosis, osteoporosis, and tumors. In transcriptional level, sterol regulating element binding protein(SREBP) and liver X receptor(LXR) are respectively responsible for regulation of lipid biosynthesis and transports, and thus play key roles in lipid metabolism. Both SREBP and LXR control signalling networks and are interconnected with each other, leading to the balance of lipid metabolism. PGC-1β is the key co-activator for SREBP, LXR and ERR?, and is of great importance in regulating lipid biosynthesis and transports.
