口服雷奈酸锶与胶原肽对尾吊大鼠骨丢失的抑制效应

【摘要】 目的观察口服雷奈酸锶(strontium ranelate,SR)与胶原肽(collagen peptides,CP)对尾吊(tail supended,TS)大鼠骨丢失的抑制效应。方法雄性SD大鼠24只随机平均分为3组:对照组(CN),尾吊模拟微重力组(TS),模拟失重同时给予SR和CP各750 mg/kg灌胃组(SR&CP),各组实验期均为28 d。采用全自动生化分析仪检测碱性磷酸酶(alkaline phosphatase,ALP)含量,放射免疫法检测骨钙素(osteocalcin,OC)含量;Micro-CT分析骨微结构;Masson染色观察骨胶原纤维;双能X线(dual energy X-ray absorptiometry,DXA)检测股骨骨密度(bone mineral density,BMD)。结果 TS大鼠补充SR和CP显示骨保护作用:抑制TS诱导的血清ALP和OC含量的降低,改善骨微结构;显著抑制TS组大鼠股骨BMD的降低。结论口服SR和CP能明显抑制TS大鼠的骨丢失,从而为微重力环境下骨丢失的防治提供了实验依据。

【Abstract】 objective To study inhibitory effect of oral administration of strontium ranelate( SR) and bovine collagen peptide( CP) on bone loss in tail suspended rats. Methods Twenty-four male Sprague Dawley( SD) rats were divided into 3 groups: normal control group( CN),tail supended group( TS),TS and once-daily intragastric administration of 750 mg / kg group( SR&CP),the experimental period was 28 d. Serum samples were analyzed for alkaline phosphatase( ALP) using an autoanalyzer; osteocalcin( OC) using radioimmunometric assay; the right femurs were analyzed for microstructure by micro CT; collagen fibers of tibia by masson staining and dual energy X-ray absorptiometry( DXA) was used to measure bone mineral density( BMD) of femur.Results TS rats supplemented with SR&CP showed osteoprotective effects: significant inhibition in reductions of their serum ALP and OC levels and femoral BMD,as well as improvement the microarchitecture of their femurs. Conclusions Oral administration SRCP can significantly inhibit bone loss of TS rats and provide theoretical basis for the prevention and treatment of osteoporosis under microgravity.
