芹菜素降低柔红霉素对小鼠心脏毒性的研究
Apigenin reduces Daunorubicin-induced cardiotoxicity in mice
【摘要】 目的研究芹菜素降低柔红霉素的心脏毒性作用及其可能的机制。方法将小鼠分为正常组、模型组及芹菜素100、200 mg·kg-1组。预先ig给药8 d后,除正常组外,其余小鼠ip给予柔红霉素30 mg·kg-1。在给药2 d后,测定小鼠血清中乳酸脱氢酶(LDH)、肌酸激酶(CK)和谷草转氨酶(AST)的活性,心肌中总超氧化物歧化酶(SOD)、谷胱甘肽还原酶(GR)、谷胱甘肽-S-转移酶、谷胱甘肽过氧化物酶(GSH-Px)的活性及心肌中丙二醛(MDA)和还原型谷胱甘肽的含量。结果与模型组小鼠相比,芹菜素处理组的小鼠血清LDH、CK、AST活性下降,而心肌组织中的GR和GSH-Px活性增加,尤以200 mg·kg-1组的作用更为明显。芹菜素也可使心肌组织中的MDA含量下降。结论芹菜素可减轻柔红霉素的心脏毒性,可能与增加心肌中GSH-Px、GR的活性而发挥的抗氧化作用有关。
【Abstract】 OBJECTIVE To investigate the effects of apigenin on the cardiotoxicity induced by daunorubicin and its potential mechanisms. METHODS The mice were randomly divided into 4 groups,namely,control group,model group,apigenin 100,200 mg·kg- 1groups. Eight days after ig administration the model and drug- treated mice were given daunorubicin 30 mg·kg- 1by ip. Whereafter,the drug- treated mice were given apigenin again for 2 days. The serum lactate dehydrogenase( LDH),creatine kinase( CK) and aspartate aminotransferase( AST) activities,the myocardial total superoxide dismutase( T- SOD),glutathione reductase( GR),glutathione S-transferase and glutathione peroxidase( GSH- Px) activities,the myocardial malondialdehyde( MDA) and reduced glutathione contents were determined. RESULTS Compared with the model mice,the serum LDH,CK and AST activities in the apigenin- treated mice were decreased,whereas the myocardial GR and GSH- Px activities were increased,especially in the apigenin 200 mg·kg- 1group.Apigenin might also decrease the myocardial MDA content. CONCLUSION Apigenin may reduce daunorubicin- inducec cardiotoxicity,may be related to its antioxidation via increment of GR and GSH- Px activities.
