新型抗银屑病药物SKLB628凝胶剂处方的筛选
Formulation screening of the gel for a novel anti-psoriasis drug SKLB628
【摘要】 目的筛选以Flt3为靶点治疗银屑病的小分子药物SKLB628的凝胶剂处方。方法通过稳定性研究来筛选4种凝胶基质,再采用透析膜扩散法对选定的处方进行体外释药试验,依据释放曲线确定最优处方。结果采用73%PEG400、12%PEG4000及15%甘油制成的凝胶基质,在耐寒耐热试验和离心试验中,外观及涂展性均无明显变化,且以其制成的SKLB628凝胶在体外的释放良好。结论最优处方是以PEG400、PEG4000等辅料为基质制得的SKLB628凝胶剂,其工艺简单,释放度良好。
【Abstract】 OBJECTIVE To select the optimal formulation of the gel for an anti- psoriasis drug SKLB628,which targets Flt3.METHODS Four gel bases were tested by stability experiment. Then semi- permeable membrane diffusion method was used to measure the in vitro releasing degrees of different formulations. According to the stripping- time curve,the best prescription was determined. RESULTS The gel bases made of 73% PEG400,12% PEG4000 and 15% glycerin showed no significant changes in the spread and appearance in the stability experiment suchas cold- resistant and heat- resistant tests. And besides,SKLB628 in this gel released well in vitro. CONCLUSION The best formulation of SKLB628 gel was PEG400- PEG4000- glycerin,which obtained simple preparation and good release.
