玉米赤霉烯酮和氯化镉对MCF-7细胞增殖的联合作用
Combined effects of zearalenone and cadmium chloride on proliferation of MCF-7 cells
【摘要】 目的研究玉米赤霉烯酮(ZEA)和氯化镉(CdCl2)联合作用对MCF-7细胞增殖的影响。方法调整MCF-7细胞密度约为1×105/mL,以0(对照)、10-8、10-7、10-6、10-5、10-4、10-3mol/L CdCl2分别染毒MCF-7细胞24h,以0(对照)、6.25、12.5、25、50、100nmol/L ZEA分别染毒MCF-7细胞24h,采用CCK-8法检测CdCl2和ZEA对MCF-7细胞增殖的影响,同时进行3×3析因设计研究CdCl2和ZEA对MCF-7细胞增殖的联合作用。结果 CdCl2和ZEA分别在10-6 mol/L、100nmol/L时促增殖效应均达到最大,相对增殖率分别为112.3%和114.6%,明显高于阴性对照组的100.0%;析因实验结果显示CdCl2和ZEA对MCF-7细胞增殖的影响表现为相加作用。结论 CdCl2和ZEA在一定剂量范围内能促进MCF-7细胞增殖,表现为明显的拟雌激素活性,二者联合作用表现为加和效用。
【Abstract】 Objective To study the combined effects of zearalenone(ZEA)and cadmium chloride(Cdcl2) on proliferation of MCF-7 cells.Methods MCF-7 cells were treated with 0,10-8,10-7,10-6,10-5,10-4 and 10-3 mol/L respectively of Cdcl2 for 24h.MCF-7cells were treated with 0,6.25,12.5,25,50 and 100nmol/L respectively of ZEA for 24 h.Cell counting kit 8(CCK8)was used to detect the effects of CdCl2 and ZEA respectively on proliferation of MCF-7cells,and the combined effects of CdCl2 and ZEA on proliferation of MCF-7cells were evaluated by 3×3factorial analysis.Results Compared with the control group,the relative proliferation rate in the group treated with 10-6 mol/L of CdCl2 and treated with 100nmol/L of ZEA was the highest,respectively was 112.3% and 114.6%.Combined effects of CdCl2 and ZEA to proliferation of MCF-7 cells for 24 hwere additive effect type by factorial experiment.Conclusion CdCl2 and ZEA in certain dose range can promote the proliferation of MCF-7,showing obvious estrogenlike activity,and the combined effects of CdCl2 and ZEA are additive effect type.
