观察电针“百会”、“涌泉”两穴对不同月龄APP/PS1双转基因小鼠海马Aβ淀粉样沉淀及超微结构的影响
Effect of electro-acupuncture intervention on hippocampus Aβ stain and ultrastructure in APP / PS1double transgenic rats
【摘要】 目的观察电针"百会"、"涌泉"两穴对5月龄和10月龄淀粉样前体蛋白/早老蛋白1(amyloid precursor protein/presenilin-1,APP/PS1)双转基因鼠海马β淀粉样蛋白(AmyloidβProtein,Aβ)沉积及超微结构的影响。方法分别将4月龄和9月龄的APP/PS1双转基因小鼠按随机分配法分为模型组、电针组和药物组,以相同月龄同窝阴性小鼠作为对照组,4月龄每组12只,9月龄每组11只。电针组取"百会"、"涌泉"两穴,每次留针时间为15分钟,隔日1次,治疗5周;药物组予多奈哌嗪片0.92 m L/g灌胃,取脑透射电镜片并观察。结果 5月龄透射电镜结果显示:在微血管、神经元突触及神经元超微结构上对照组优于模型组。10月龄透射电镜结果显示:模型组海马出现Aβ淀粉样沉淀——老年斑。结论 5月龄小鼠行为学出现阿尔茨海默病样改变;10月龄APP/PS1双转基因小鼠海马出现老年斑;电针治疗APP/PS1双转基因小鼠可能对海马超微结构有保护作用。
【Abstract】 Objective To observe the influence of electro-acupuncture( EA) at"Baihui"( GV20)and "Yongquan"( KI1) on hippocampal Aβ stain and ultrastructure in 5-month-old and 9-month-old APP /PS1 double transgenic rat. Methods 4-moth-old and 9-month-old APP / PS1 double transgenic rats were according to the random assignment method divided into model group,EA group and drug( ACh E) group and drug group with 12 rats each in 4-month-old groups and 11 rats each in 9-month-old groups,and the same months old rats with brood recessive gene were set as control group. Treatment was applied to"Baihui"( GV20) and"Yongquan"( KI1) for 15 mins,once every 2 days for 5weeks; For the drug group,0. 92 m L / g of acetylcholine enzyme was given by gavage,once a day. Rat brains were taken to make transmission electric lens for observation. Results On transmission electron microscopy( sem) results showed that in 5-month-old rats,the capillaries,synapses and ultrastructure of neurons in the control group were better than the model group. In 10-month-old rats,there was senile plaque in the model group. Conclusion Alzheimer’s disease-like behavior appeared in 5-month-old rats. Senile plague in hippocampus appeared in10-month-old APP / PS1 double transgenic rats. EA therapy might improve the hippocampus microstructure in APP / PS1 double transgenic rats.
