东北小麦白粉菌遗传多样性及其地域关联性分析

【摘要】 利用ISSR分子标记技术,分析从辽宁春麦区及山东、河南、湖北冬麦区收集并鉴定的26个小麦白粉菌生理小种的遗传多态性,并探讨菌株遗传多态性与其来源地的关联性。用18条ISSR引物对各小种的DNA进行扩增,其中8条引物能产生稳定的多态性图谱。多态性聚类分析结果显示,26个小麦白粉菌菌株可聚为4类,菌株的DNA多态性与其毒性多态性之间存在一定程度的关联性。依据ISSR聚类分析结果,辽宁春麦区和其他3省的优势小种、次优势小种按相应类群聚在一起,尤其是主要小种11号、411号与近邻山东冬麦区的小种聚为一类。说明东北春麦区小麦白粉病菌与南部冬麦区尤其是山东的小麦白粉病菌具有较高的亲缘关系,为东北春麦区白粉病初菌源来自山东等南部冬麦区的早期推论提供了DNA分子佐证。

【Abstract】 The genetic diversity of 26 Blumeria graminis f. sp. tritici races collected from Liaoning spring wheat region and Shandong,Henan and Hubei winter wheat regions was analyzed by ISSR( inter-simple sequence repeat)-PCR,and the relationship with their geographic sources was also explored. During 18 random primers used,eight primers could produce stable DNA polymorphism map. According to the DNA polymorphism cluster analysis,26 races clustered into four groups. To some extent,the correlation existed between virulence polymorphism and DNA polymorphism of Blumeria graminis from above four provinces.Based on the DNA polymorphism cluster analysis result,the most predominant races and the subpredominant races from the above regions could come together into same clusters,especially the most predominant races 11 and 411 of Liaoning and Shandong provinces. The above molecular evidence indicated a high phylogenetic relationship between the population of Blumeria graminis in northeast spring wheat region and those in Hubei,Henan,especially Shandong winter wheat regions,supporting the early speculation that the initial inoculum source of wheat powdery mildew in northeast spring wheat region comes from Shandong province and other southern winter wheat regions.
