硫酸锰浸种对黑柴胡种子萌发及抗氧化酶活性的影响
Effects of MnSO4 Soaking on Germination and Antioxidant Enzyme Activities of Bupleurum smithii Wolff. Seeds
【摘要】 为研究硫酸锰对黑柴胡种子萌发的影响及其生理机制,促进黑柴胡种子萌发,采用0.25~1.00 g/L的硫酸锰溶液对黑柴胡种子进行浸种,研究种子发芽速度、发芽势、发芽率及种子萌发过程中抗氧化酶活性和丙二醛(MDA)含量的变化。结果表明,硫酸锰处理可缩短种子萌发启动时间,并提高种子发芽率和发芽势,其中以0.50 g/L处理效果最好。与清水对照相比,0.50 g/L硫酸锰处理后黑柴胡种子MDA含量降低,超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性升高,这表明硫酸锰浸种处理能提高种子萌发期间的抗氧化水平,减轻膜脂过氧化作用,从而维持种子质膜结构完整性,促进种子萌发。
【Abstract】 In order to study the effect of Mn SO4 on germination of Bupleurum smithii Wolff. seeds and its physiological mechanisms,Bupleurum smithii Wolff. seeds were soaked by 0. 25—1. 00 g / L Mn SO4,the germination speed,germination potential,germination rate,activities of antioxidant enzymes and the content of MDA during seed germination were determined. The results showed that the germination starting time of Bupleurum Smithii Wolff. seeds treated by Mn SO4 was shortened while the seeds germination rate and germination potential increased,the optimum concentration was 0. 50 g / L. Compared with the control without Mn SO4,the MDA content decreased while the activities of superoxide dismutase( SOD),peroxidase( POD) and catalase( CAT) increased after Mn SO4 soaking. The results indicated that Mn SO4 treatment could enhance antioxidation level,mitigate the membrane lipid peroxidation,maintain the integrity of plasma membrane,and accelerate the seeds germination.
