利用ISSR分子标记鉴定苦瓜杂交种纯度(英文)

【摘要】 [目的]采用ISSR分子标记对苦瓜杂交一代品种秀玉1号纯度进行鉴定,为生产上提高苦瓜新品种纯度鉴定效率提供参考。[方法]利用ISSR技术和91条引物,对苦瓜品种秀玉1号及其亲本的DNA指纹进行分析。[结果]从91条引物中筛选出两条特征引物能同时扩增出F1代及其父母本的多态性条带,分别是ISSR-845和ISSR-891,两个特异位点分别位于510和300 bp处。其中ISSR-891能够准确区分母本自交系与杂种F1,经群体验证表明该标记与田间鉴定结果高度一致。[结论]ISSR技术具有准确、简便、高效和实用强等优点,可用于生产上苦瓜种子纯度的快速鉴定。

【Abstract】 [Objective] This study was conducted to verify the feasibility of ISSR marker for identifying genetic purity of bitter gourd hybrid, and thus to provide an effective method for seed purity test in production practices. [Method] The DNA fingerprints of a bitter gourd cultivar Xiuyu 1 and its parents were analyzed using ISSR marker with 91 primers. [Result] Two primers ISSR-845 and ISSR-891 which amplified two DNA bands of 510 and 300 bp respectively from F1 generation and its parents were screened out from 91 primers. ISSR-845 could distinguish the male parent from F1 hybrid and the female parent, while ISSR-891 could distinguish the female parent inbred line from Xiuyu 1. Seed purity test with the specific markers gave the same result with filed trials based on morphological identification. [Conclusion] ISSR marker is an accurate, simple and effective method for seed purity test bitter gourd hybrid, and thus can be used in production practices.
