羟基酪醇和橄榄苦苷对过氧化氢诱导的猪肾小管上皮细胞氧化损伤的拮抗作用
Antagonism of hydroxytyrosol and oleuropein on the oxidative damage of procine LLC-PK1 cells caused by hydrogen peroxide
【摘要】 目的探讨羟基酪醇和橄榄苦苷在氧化应激致猪LLC-PK1(p LLC-PK1)细胞损伤中的拮抗作用。方法用10200μmol/L过氧化氢处理pLLC-PK1细胞,筛选最佳的处理时间与浓度,建立LLC-PK1细胞过氧化氢氧化应激体外模型。LLC-PK1细胞分别与2、5和10μmol的羟基酪醇和橄榄苦苷孵育12 h,用100μmol/L的过氧化氢作用4 h,裂解LLC-PK1细胞后,检测丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力和谷胱甘肽过氧化物酶(GSH-Px)活力和脂肪酸组成变化。结果与过氧化氢模型组(100μmol/L)相比,在橄榄苦苷和羟基酪醇处理的LLC-PK1细胞中,MDA表达量降低,SOD和GSH-Px活力显著升高,脂肪酸转化降低,差异均有统计学意义(P<0.05,P<0.01)。结论羟基酪醇和橄榄苦苷对过氧化氢诱导的LLC-PK1细胞氧化应激具有拮抗作用。
【Abstract】 Objective To explore the antagonism of hydroxytyrosol and oleuropein on porcine LLC-PK1 cells caused by hydrogen peroxide(H2O2). Methods The in vitro oxidative stress model of the LLC-PK1 cells was established based on the opitimized time and concentration of the H2O2 treatment, cell viability was tested by MTT assay. After the LLC-PK1 cells were incubated with 2 μmol/L, 5 μmol/L and 10 μmol/L of hydroxytyrosol and oleuropein for 12 h,respectively,100 μmol/L of H2O2 was added into the treated LLC-PK1 cells,incubating for 4 h. After the LLC-PK1 cells were lysed,the MDA content,SOD activity,GSH-Px activity,and fatty acid transformation were detected,respectively. Results Compared with H2O2 model group,in the LLC-PK1 cell groups treated by hydroxytyrosol and oleuropein,the MDA content decreased significantly,the activities of both SOD and GSH-Px exhibited a significant increase,and the fatty acid transformation showed a significant decrease.Conclusion Hydroxytyrosol and oleuropein have a potential preventive effect on LLC-PK1 cells suffering from the damage by H2O2 oxidative stress.
