铅对小鼠睾丸细胞活性和活性氧的影响

【摘要】 目的探讨乙酸铅对小鼠睾丸细胞活性和活性氧(ROS)水平的影响。方法将具有小鼠原代睾丸支持细胞特性的TM4细胞株暴露于不同浓度乙酸铅溶液中24 h,采用新型四唑盐WST-8方法检测乙酸铅诱导的细胞增殖情况;采用流式细胞术检测细胞内ROS水平变化。结果与对照组相比,低浓度铅诱导细胞增殖,随着乙酸铅浓度升高,细胞存活率逐渐降低(P<0.05),呈现先增高后降低的剂量-效应关系。ROS结果显示在一定剂量范围内,随着乙酸铅浓度的升高,细胞内ROS含量显著升高(P<0.05)。结论低剂量乙酸铅对小鼠睾丸细胞有兴奋效应,高剂量会抑制细胞增殖,同时可诱导氧化应激反应,引起细胞损伤。

【Abstract】 Objective To explore the effects of lead on the cellular activity and ROS levels in testicle cells of mice. Methods TM4 cell strain with the characteristics of primary Sertoli cells were exposed to different concentrations of lead acetate for 24 hours,the cell proliferation was measured by WST-8 assay and the reactive oxygen toxicity(ROS) levels was detected by flow cytometry. Results Compared with the control group,the viability of TM4 cells had a trend to increase when the concentration of lead acetate was lower,then the cell viability started to decline(P <0.05). ROS levels increased significantly in a dose-dependent manner in the cells treated by lead acetate within a certain concentration range( P<0.05). Conclusion Low does of lead acetate may cause hormesis on testicle cells of mice,high does can inhibit cell proliferation and induce oxidative stress,causing cell damage.
