二苯甲酮-3对绒毛外滋养层细胞的增殖及miR-17-92基因簇表达的影响
Effects of benzophenone-3 on cell proliferation and expression of miR-17-92 cluster mRNA in HTR-8/Svneo cells
【摘要】 目的研究二苯甲酮-3(benzophenone-3,BP-3)对绒毛外滋养层(HTR-8/Svneo)细胞的增殖及miR-17-92基因簇表达的影响。方法将HTR-8/Svneo细胞分别暴露于含终浓度0(溶剂对照)、0.01、0.1、1 mg/L BP-3的培养基中培养24 h。采用CCK-8法检测细胞活性,采用流式细胞仪检测细胞凋亡情况,采用Real-time PCR方法检测细胞miR-17-92基因簇的表达水平。结果与溶剂对照组相比,1 mg/L BP-3染毒组HTR-8/Svneo细胞的存活率和miR-17、miR-19a mRNA的表达水平均降低,差异有统计学意义(P<0.05);而各剂量BP-3染毒组HTR-8/Svneo细胞的凋亡率及miR-18a、miR-19b、miR-20a、miR-92a mRNA的表达水平均无明显变化。且随着BP-3染毒剂量的升高,HTR-8/Svneo细胞的存活率呈下降趋势,凋亡率呈上升趋势。与抑制剂对照组相比,抑制miR-17和miR-19a表达后HTR-8/Svneo细胞存活率均降低,差异均有统计学意义(P<0.05)。结论 BP-3可通过抑制miR-17-92簇中miR-17、miR-19a的表达,抑制HTR-8/Svneo细胞增殖。
【Abstract】 Objective To investigate the effects of benzophenone-3(BP-3) on HTR-8/Svneo cell proliferation and the expression of miR-17-92 cluster m RNA. Methods HTR-8/Svneo cells were respectively exposed to the medium with the final concentrations of 0(with solvent as the control),0.01,0.1 and 1 mg/L BP-3 and then cultured for 24 h. Cell viability was detected by CCK-8, cell apoptosis was detected by flow cytometry, and the relative expression level of miR-17-92 cluster mRNA was detected by real-time PCR method. Results Compared with the solvent control group,the survival rate of HTR-8/Svneo cells in the 1 mg/L BP-3 exposure group was lower,and the expression level of miR-17 and miR-19 a mRNA of HTR-8/Svneo cells was also lower, with statistically significant differences(P <0.05). The apoptosis rates and the expression level of miR-18 a, miR-19 b, miR-20 a and miR-92 a mRNA of HTR-8/Svneo cells showed no significant differences in all BP-3exposure groups. With the increase of the BP-3 dose,the survival rate of HTR-8/Svneo cells showed a decreasing trend,and the apoptosis rate showed an increasing trend. Compared with the inhibitor control group,the survival rate of HTR-8/Svneo cells in the miR-17 and miR-19 a inhibitor group was lower, with statistically significant differences( P <0.05). Conclusion BP-3 can inhibit the HTR-8/Svneo cell proliferation through reducing the expression level of miR-17-92 cluster miR-17 and miR-19 a.
