低温体外保存对血小板活化和凋亡的影响
Effects of low temperaure on platelet activation and apoptosis
【摘要】 目的观察保存温度对血小板活化和凋亡的影响,评估低温体外保存血小板的效果。方法取20份单采血小板,一分为三,一份于(22±2)℃振荡保存(对照组),另两份悬浮于血小板添加液(65%)与自体血浆(35%)的混合液中分别静置于10℃储血冰箱(实验Ⅰ组)和(22±2)℃振荡仪(实验Ⅱ组),于1、3、4、5、7d检测血小板计数(platelet count,PLT)、血小板P选择素(cluster of differentiation 62platelet,CD62p)表达率、血小板线粒体跨膜电位(mitochondrial transmembrane potentia,△Ψm)。结果随着时间延长,3组PLT比较平稳,在组间、时点间以及组间·时点间交互作用差异均无统计学意义(P>0.05);随着时间延长CD62p逐渐升高,而△Ψm%逐渐下降,3组CD62p、△Ψm在组间、时点间以及组间·时点间交互作用差异均有统计学意义(P<0.01)。结论血小板添加液混合血浆10℃低温保存比(22±2)℃保存血小板的活化和凋亡程度均较低。
【Abstract】 Objective To study the effects of low temperature on platelet activation and apoptosis.Methods Twenty parts of apheresis platelets were divided into three groups,one group for oscillation(control group),and the other two groups saved in the liquid with platelet preservation solution(65%)and autologous blood plasma(35%)under 10 ℃ refrigerator(experiment groupⅠ)and(22±2)℃ oscillator(experiment groupⅡ).Platelet count(PLT),cluster of differentiation 62platelet(CD62p),mitochondrial transmembrane potential(△Ψm)on1d,3d,4d,5dand 7dwere tested.Results PLT of three groups was smooth with extended time.There were no significant difference between three groups(P>0.05).With prolonged time,CD62 p gradually rose,but △Ψm decreased significantly(P<0.01).CD62 p and △Ψm of three groups had no significant difference on 1-3d(P>0.05);CD62p of the experimental Ⅱ was above the control group and the experiment groupⅠ on 5dand 7d(all P<0.01),but there were no significant difference between the group experimentalⅠ and the control(P>0.05);△Ψm of the experimental groupⅠ was significantly higher than that of the control group and the experiment groupⅡ on 4-7d(all P<0.01).Conclusion The effects of platelet activation and apoptosis in the liquid with platelets preservation solution(65%)and autologous blood plasma(35%)under 10 ℃refrigerator were better than in(22±2)℃ oscillator.
