Cyclin D1-siRNA促进口腔鳞癌Tca8113细胞凋亡
Induction of Cyclin D1 on Apoptosis of OSCC Tca8113 Cell Line
【摘要】 目的:研究细胞周期蛋白D1(cyclin D1)siRNA对口腔鳞癌细胞Tca8113凋亡的作用。方法:采用cyclin D1-siRNA慢病毒感染Tca8113,qPCR检测cyclin D1表达,MTT检测细胞活性,Annexin-V单染检测细胞凋亡。结果:Cyclin D1-siRNA慢病毒成功抑制Tca8113中cyclin D1mRNA表达。Cyclin D1干扰组细胞活性显著低于阴性病毒组(P<0.01),且细胞凋亡率高于阴性对照组(P<0.01)。结论:Cyclin D1干扰能促进Tca8113凋亡,起到抗肿瘤效应。
【Abstract】 Objective:To detecte the effect of cyclin D1 on OSCC apoptosis.Methods:Tca8113 was transduced by Lentivirus cyclin D1 siRNA to find cyclin D1 expression interrupting cell line;qPCR was used to detect cyclin D1 mRNA expression;MTT was used to detect cell viabibility and Annexin-V staining was used to detect the ratio of apoptosis.Results:Cyclin D1 expression was interrupted by Lentivirus cyclin D1 siRNA.The cell viability of cyclin D1 siRNA group decreased when compared with LV-negative group(P<0.01)and the ratio of apoptosis showed an increase(P<0.01).Conclusion:Cyclin D1 siRNA can induce apoptosis of Tca8113,and displays an anti-tumor effect.
