【摘要】 目的:探讨高效液相色谱法(HPLC)与酶法检测血清游离脂肪酸(FFA)的结果有无差异。方法:收取武汉大学中南医院92名健康体检者空腹外周血3ml,分离新鲜血清,采用全自动生化分析仪酶法测定血清FFA水平,同时以正十七酸(C17∶0)为内标,改良Doles法萃取血清FFA,HPLC法定量检测。结果:酶法检测血清FFA结果为466.5(347.3,585.3)μmol/L,HPLC法FFA结果为448.5(339.5,571.9)μmol/L,两种方法检测结果无明显差异(t=1.394,P>0.05),且具有良好的相关性(r=0.980,P<0.05)。结论:酶法与HPLC法检测FFA结果一致性良好。酶法操作简单,更适合临床实验室常规检测。更多还原

【Abstract】 Objective:To explore whether serum free fatty acids(FFA)level determined by high performance liquid chromatography(HPLC)differed from the results of enzymatic methods.Methods:A total of 92 subjects were recruited from Zhongnan Hospital of Wuhan University,3ml fasting blood samples were taken from the participants,and serum FFA levels were measured both by automatic biochemical analyzer(enzymatic method)and HPLC method.The modified Doles Extraction method was used to extract free fatty acids,and heptadecanoic acid(C17∶0)served as an internal standard.Results:There was no significant difference(t=1.394,P>0.05)between the results of enzymatic method(466.5[347.3,585.3]μmol/L)and HPLC method(448.5[339.5,571.9]μmol/L),and the results showed strongly correlation(r=0.980,P<0.05).Conclusion:The results of enzymatic method are consistant with HPLC.Enzymatic assay is simple and suitable for clinical laboratory routine testing.更多还原
