桔梗高效离体再生体系的建立
Establishment of A Highly Efficient in Vitro Regeneration System for Platycodon grandiflorum(Jacq.)A.DC.
【摘要】 分析了消毒时间、6-BA浓度、外植体放置方式、叶龄对桔梗[Platycodon grandiflorum(Jacq.)A.DC.]丛生芽形成过程的影响。结果表明,0.1%升汞消毒5 min的桔梗种子成活率最高;诱导桔梗丛生芽的最适培养基为MS+0.5 mg/L NAA+1.0 mg/L 6-BA;叶片是最佳外植体;叶片近轴面贴近培养基时,出芽快;叶龄越短,脱分化所需时间越短。
【Abstract】 The effects of disinfection time,6-BA level,e xplant orientation and leaf ages on formation of multiple shoots of Platycodon grandiflorum(Jacq.)A.DC. were examined. The results showed that the survival rate of seeds was highest under0.1% Hg Cl2 disinfection for 5 minutes. The best medium for multiple shoots induction was MS+0.5 mg/L NAA+1.0 mg/L 6-BA and the best explant was leaf. When it contact the medium, the adaxial surface of leaf explant was better for shoot induction.The younger a leaf age was, the sooner the leaf dedifferentiated.
