茯苓多糖对人胃癌裸鼠移植瘤的抑制效应研究
Inhibitory Effect of Pachyman on Human Gastric Cancer of Xenografts in Nude Mice
【摘要】 目的探讨茯苓多糖对人胃癌裸鼠移植瘤的抑制效应及部分作用机制。方法用体外培养的SGC-37901人胃癌细胞株制作皮下荷瘤裸鼠模型40只,随机分为5组:茯苓多糖低、中、高剂量组[予茯苓多糖400、200、100 mg/(kg·d)],环磷酰胺组[予环磷酰胺20 mg/(kg·d)],对照组(予相同体积生理盐水),每组8只。均予灌胃给药。观察各组肿瘤重量的变化并计算抑瘤率及肝、脾指数;采用免疫组化染色法研究肿瘤组织的凋亡情况以及凋亡相关基因Bax及Bcl-2表达的变化。结果与对照组比较,用药各组的瘤质量均显著降低(P<0.05);与环磷酰胺组比较,茯苓多糖低、中、高剂量组的瘤质量、抑瘤率差异均有统计学意义(P<0.05),且随剂量的增加抑瘤作用显著增加(P<0.05)。茯苓多糖各组脾指数明显高于对照组与环磷酰胺组(P<0.05),肝指数均低于对照组与环磷酰胺组(P<0.05)。环磷酰胺组、茯苓多糖各组与对照组Bcl-2蛋白阳性指数(positive index,PI)比较下降明显(P<0.05);茯苓多糖各组Bcl-2蛋白PI均明显高于环磷酰胺组(P<0.05);茯苓多糖中、高剂量组与低剂量组比较,Bcl-2蛋白PI下降明显(P<0.05),中、高剂量组差异不明显(P>0.05)。环磷酰胺组、茯苓多糖各组与对照组Bax蛋白PI比较升高明显(P<0.05);茯苓多糖各组Bax蛋白PI均明显低于环磷酰胺组(P<0.05),茯苓多糖各组随浓度增加,Bax蛋白PI显著升高(P<0.05)。结论茯苓多糖对裸鼠胃癌原位移植瘤具有抑制作用,其机制可能是通过提高机体免疫力的同时调控Bcl-2/Bax蛋白,促进肿瘤细胞凋亡。
【Abstract】 Objective To investigate the inhibitory effect and some mechanisms of Pachyman on human gastric cancer of xenografts in nude mice. Methods A total of 40 nude mouse models of subcutaneous bearing cancer were established by culture in vitro with SGC-7901 human gastric cancer of cell line,and were randomly divided into low,middle and high doses of Pachyman groups [n = 8,Pachyman 100,200 and 400 mg /( kg·d) ],Cyclophosphamide group[n = 8,Cyclophosphamide 200 mg/( kg·d) ]and control group( n = 8,same volume of physiological saline),and all groups underwent intragastric administration. Changes of tumor masses were observed,and inhibitory rates of tumor and the liver and spleen indexes were calculated in all groups. The apoptosis of tumor tissues and the changes of Bax and Bcl-2 expressions were analyzed using immunohistochemical staining method. Results The tumor masses in medication groups were significantly reduced compared with that in control group( P < 0. 05); the differences in tumor masses and inhibitory rates of tumor were statistically significant in low,middle and high doses of Pachyman groups compared with that in Cyclophosphamide group( P < 0. 05),and the inhibitory effect enhanced with increasing dose( P < 0. 05). The values of spleen indexes in Pachyman groups were significantly higher( P < 0. 05),while the values of liver indexes were significantly lower than those in Cyclophosphamide and control groups( P < 0. 05). The positive indexes( PI) of Bcl-2protein in Pachyman groups and Cyclophosphamide group were significantly decreased compared with that in control group( P < 0. 05); the PI values of Bcl-2 protein in Pachyman groups were significantly higher than that in Cyclophosphamide group( P < 0. 05); PI values of Bcl-2 protein in the middle and high doses Pachyman groups were significantly decreased compared with that in the low dose group( P < 0. 05),but the difference in the middle and high doses groups was not significant( P > 0. 05). PI values of Bax protein in Pachyman groups and Cyclophosphamide group were significantly increased compared with that in the control group( P < 0. 05); PI values of Bax protein in Pachyman groups were significantly lower than that in Cyclophosphamide group( P < 0. 05),and the PI values of Bax protein were increased with the increasing Pachyman concentration in Pachyman groups( P < 0. 05). Conclusion Pachyman has inhibitory effect on orthotopic transplantation tumor of gastric carcinoma in nude mice,and the mechanisms may be obtained by improving the immune system and regulating the Bcl-2 / Bax protein at the same time so as to promote apoptosis of tumor cell.
