苦参碱联合卡铂对人胃癌SGC-7901细胞增殖及凋亡的影响
Effects of matrine and carboplatin of proliferation and apoptosis in human gastric carcinoma SGC-7901 cells
【摘要】 目的探讨苦参碱联合卡铂对人胃癌SGC-7901细胞抑制增殖及诱导凋亡的机制。方法将体外培养人SGC-7901细胞分为对照组、苦参碱组(1.0mg/m L、2.0mg/m L、3.0mg/m L)、卡铂组(0.01mg/m L)及苦参碱联合卡铂组。MTT检测各组细胞增殖抑制率,FCM检测各组细胞凋亡率,RT-PCR检测各组细胞的CXCR4m RNA表达。结果相同浓度苦参碱联合卡铂组增殖抑制率及凋亡率明显高于单药苦参碱组(P<0.05)及其单药卡铂组(P<0.05)。与对照组比较,各实验组CXCR4m RNA灰度值均降低(P<0.05)。相同浓度苦参碱联合卡铂组灰度值明显低于单药苦参碱组(P<0.05)及其单药卡铂组(P<0.05)。结论苦参碱联合卡铂能发挥正协同作用人胃癌SGC-7901细胞,剂量依赖性的抑制其增殖并诱导凋亡,该作用可能与抑制细胞内CXCR4 m RNA表达有关。
【Abstract】 Objective To investigate the effects of matrine and carboplatin on the CXCR4 expression of proliferation and apoptosis in human gastric carcinoma SGC-7901 in vitro,and its potential mechanism. Methods SGC-7901 cells were treated with matrine(final concentrations=1.0,2.0 and 3.0mg/m L) and carboplatin(final concentrations=0.01mg/m L),either each alone or in combination.The viability in treated SGC-7901 cells was assessed by MTT.Cell apoptosis of SGC-7901 was assessed by flow cytometry.CXCR4 m RNA abundance by RT-PCR. Results Matrine and carboplatin,either each alone or in combination,significantly increased inhibition rate(P < 0.05),significantly higher apoptotic rate(P < 0.05),and decreased CXCR4 m RNA abundance(P < 0.05).The above effects of SGC-7901 cells were more pronounced when they were jointed in combination than they were used alone(P < 0.05). Conclusion Matrine and carboplatin in combination can signifcantly inhibit the proliferation and induce apoptosis of SGC-7901 cells in a dose-dependent manner,the observed effects of matrine may be related to decreased CXCR4 expression.
