一株产赖氨酸脱羧酶的菌株Klebsiella pneumoniae sp.GXW-1的鉴定
Identification of a Klebsiella pneumoniae sp. GXW-1 which Produced Lysine Decarboxylase
【摘要】 已有研究表明尸胺可以代替己二胺和己二酸合成新型尼龙,从而解决工业上用石油制取己二胺带来的资源枯竭及环境污染问题,并且尸胺是赖氨酸脱羧酶的产物。本研究通过微生物纯培养技术筛选得到了一株产赖氨酸脱羧酶的菌株GXW-1,完成了该菌株16S rDNA基因鉴定和生理生化特性研究,将其鉴定为Klebsiella pneumoniae sp.。该菌种酶活的最适作用温度为40℃,最适p H为5.4,其酶活为80.46 U,具有较好的生化特性优势。研究表明,金属离子Cd2+和Sr2+可以刺激酶活的提升,而Trition试剂对酶活性也有微弱的激活作用。本研究为以后的工业化生产尸胺提供了思路和依据。
【Abstract】 Studies have shown that cadaverine can replace hexamethylene diamine and adipic acid by synthesizing new synthetic nylon, so as to solve the problem of resource depletion and environmental pollution brought by industrial oil preparating hexamethylene diamine, and cadaverine is the production of lysine decarboxylase. This study screened a Klebsiella pneumoniae sp. GXW-1 strain which could produce L-lysine decarboxylase by microbial pure culture technology and finished the identification of 16 S r DNA gene and characterization of physiology and biochemistry. Its enzymatic activity was 80.46 U under the optimal reactions of the temperature 40℃ and p H 5.4. It had better biochemical characterization compared with the other similar microorganisms. Further study revealed that metal ions of Cd2+and Sr2+could stimulate the enzymatic activity, and Trition reagent also had the similar activation. This research provides tactics and some bases for industrial production of cadaverine.
