【摘要】 目的研究基质金属蛋白酶-9(MMP-9)蛋白及mRNA在乳腺癌中的表达,探讨其在乳腺癌发生、发展及转移过程中的临床意义。方法用免疫组化法检测MMP-9蛋白在乳腺癌、乳腺良性病变组织中的表达,用半定量反转录聚合酶链反应(RTPCR)的方法检测组织中mRNA的表达。结果 MMP-9蛋白在乳腺癌组织中的阳性表达率为69.6%(39/56),在乳腺良性病变组织中为20%(6/30),差异有统计学意义(P<0.05)。MMP-9mRNA在乳腺癌和乳腺良性病变组织中表达水平(灰度比值)分别为0.914 2±0.108 1、0.379 4±0.042 8,两者比较差异有统计学意义(P<0.05)。结论 MMP-9蛋白及mRNA在乳腺癌组织中的表达与淋巴结转移呈正相关,有可能作为判断乳腺癌浸润转移能力的参考指标之一。更多还原

【Abstract】 Objective To study expression of matrix metalloproteinase-9(MMP-9)protein and its mRNA in breast cancer and evaluate its significance in the occurrence,development and metastasis of breast cancer.Methods The protein expression of MMP-9breast cancer were detected by using immunohistochemistry and the expression of mRNA were detected by reverse transcription polymerase chain reaction(RT-PCR)Results The positive rate of MMP-9protein expression in 56 cases of breast cancer was69.6%(39/56),while in benign breast diseases was 20%(6/30),which were significantly different(P<0.05).The levels of MMP-9mRNA were significantly higher in the patients with breast cancer than those in benign breast diseases(P<0.05),which were0.914 2±0.108 1and 0.379 4±0.0428 respectively.Conclusion The MMP-9protein and mRNA expression in human breast cancers are positively correlated with the stage and lymph node metastasis.Expression of MMP-9could be used as an indicator for the ability of invasion and metastasis of breast cancer.更多还原
