利用Image J软件对家兔动脉粥样硬化病变图像进行定量分析的方法

【摘要】 目的动脉粥样硬化是心血管病的病理基础之一,而心血管病是全世界最主要的疾病致死原因。许多成熟的动物模型能够很好的模拟人类动脉粥样硬化的复杂病变,极大地促进了动脉粥样硬化的研究。然而,这些复杂病变也增加了研究者的分析难度。为此我们提出一套行之有效的图像分析方法对这些病变进行分析,并且该方法基于跨平台的免费图像软件-Image J。方法首先,苏丹Ⅳ和EVG(Elastic Van Gieson)染色被用来估算动脉粥样硬化病变的大体面积和镜下面积。其次,通过对免疫组化图像进行分析,病变中的细胞成分被准确定量。最后,家兔心脏被切成7个包块以便于在较大的范围寻找和分析冠脉动脉粥样硬化病变。结果我们提出的这套动脉粥样硬化病变的图像分析方法,能够准确和有效的评估动脉粥样硬化病变程度。结论该研究方法降低了研究工作难度和研究成本,并能够推广到更多的动物模型及临床组织学图像的分析中去。

【Abstract】 Objective Atherosclerosis is regarded as a basic pathogenesis of cardiovascular diseases,which are the leading cause of mortality worldwide.Sophisticated animal models radically improve the research of atherosclerosis because these animal models recapitulate the complicated pathogenesis of human atherosclerosis.However,these animal models also present a challenge to analyzing these complicated lesions accurately and efficiently.In the current study,we present a suit of practical methods to analyze the complicated lesions in animal models,based on a cross-platform free software Image J.Methods First,we assessed the size of gross and microscopic lesions by SudanⅣ and EVG(Elastic van Gieson)staining respectively.Second,cellular components in lesions were identified by Image J via spitting the RGB image of immunohistochemistry(IHC)to monochromatic image for exclusion of background.Moreover,and we developed a method to analyze the coronary atherosclerosis in the whole rabbit heart by seperating the heart into 7blocks.Results The atherosclerosis in rabbits was accurately and comprehensively revealed by ours methods.Conclusion These methods will be widely introduced for analysis in other animal models and clinical invisitigation.
