Sox9启动子构建及与其与Ⅱ型胶原蛋白基因调控关系的实验研究

【摘要】 目的构建Sox9启动子观察其与软骨相关基因Sox9和Ⅱ型胶原蛋白基因的调控与影响。方法构建和鉴定Sox9启动子,观察其在人体软骨肉瘤细胞中对Sox9基因和Ⅱ型胶原蛋白基因COL2A1 mRNA表达的影响。结果测序结果与设计的Sox9启动子转录模板序列完全相同,Sox9启动子转染的人体软骨肉瘤细胞中Sox9基因和Ⅱ型胶原蛋白基因COL2A1的mRNA表达增加。结论构建Sox9启动子具有转录功能,可以稳定转染人体软骨肉瘤细胞,转染后可以增加Sox9基因和Ⅱ型胶原蛋白基因COL2A1 mRNA的表达。

【Abstract】 Objective To construct the Sox9 promoter and observe its gene regulation of Sox9 and collagen Ⅱ gene COL2A1. Methods Sox9 promoter was constructed and identified, and its gene regulation of Sox9 and collagen Ⅱ gene COL2A1 in human chondrosarcoma cells was observed. Results After Sox9 promoter was transferred in chondrosarcoma cells the expressions of Sox9 and COL2A1 m RNA were increased compared with the control. Conclusion The constructed Sox9 promoter has transfer function and plasmid could be transferred into human condrosarcoma cells. It could increase the expression of Sox9 and COL2A1 genes.
