衰老对内皮细胞膜微粒及内皮细胞功能的影响

【摘要】 目的探讨衰老对内皮细胞膜微粒(EMVs)及内皮细胞功能的影响。方法年轻(yEMVs)和年老EMVs(aEMVs)分别来源于年轻(3~5代)、年老(25~28代)人脑微血管内皮细胞(HBMECs)。年轻与老年HBMECs与yEMVs或aEMVs共培养,检测miR-34a表达、细胞增殖、血管生成能力及活性氧、一氧化氮水平。结果老年HBMECs释放的EMVs中miR-34a表达明显高于年轻HBMECs。aEMVs增加HBMECs表达miR-34a及活性氧生成,但抑制细胞增殖、血管生成及一氧化氮产生;yEMVs则有相反作用。结论衰老引起内皮细胞释放不同EMVs,影响内皮细胞功能,并且可能与EMVs所携带的miR-34a有关。

【Abstract】 Objective To investigate the effect of aging on the endothelial cell-derived microvesicles(EMVs) and endothelial function. Methods The young(y) and aged(a) EMVs were respectively derived from young(3-5 passages) and aged(25-28 passages) human brain microvascular endothelial cells(HBMECs). After co-cultured with y EMVs or a EMVs, miR-34 a expression, proliferation, angiogenesis and levels of nitric oxide(NO) and reactive oxygen species(ROS) were determined in the young and aged HBMECs. Results miR-34 a expression was upregulated in a EMVs compared with yEMVs. The aEMVs increased miR-34 a expression and reactive oxygen sepacies production, but decreased cell proliferation, angiogenesis, and production of nitric oxide and reactive oxygen species in HBMECs. However, yEMVs exhibited the opposing effects in HBMECs. Conclusion Aging can induce the release of different EMVs and affect the function of HBMECs, which might be related to miR-34 a expression in EMVs.
