当归多糖对小鼠酒精性肝细胞损伤的作用
The protection mechanism research of Angelica sinensis polysaccharide on alcoholic liver injury in mice
【摘要】 目的 探讨当归多糖对小鼠酒精性肝细胞损伤的保护机制。方法 1通过一次性高浓度酒精灌胃建立小鼠急性酒精性肝损伤模型,之后将小鼠随机分为当归多糖组、酒精模型组,同时设正常对照组。当归多糖组连续腹腔注射当归多糖6 d,酒精模型组及正常对照组腹腔注射等量生理盐水。动态观察小鼠的一般情况、肝组织形态学结构变化。2对各组血清丙二醛、超氧化物歧化酶及谷胱甘肽过氧化物酶进行生化检测。结果 1小鼠酒精性肝损伤腹腔注射当归多糖组,肝组织逐渐得到明显修复,约11 d肝小叶完全修复正常;而酒精性肝损伤未注射当归多糖组,肝组织修复差;正常对照组,肝组织结构无变化。2血清生化检测显示,比较当归多糖组与酒精模型组血清丙二醛、超氧化物歧化酶及谷胱甘肽过氧化物酶生化检测有统计学意义。结论 当归多糖可通过提高抗氧化酶活性,清除氧自由基,减轻膜脂质过氧化链式反应,继而使肝细胞的生物膜结构恢复,从而对肝细胞起到保护作用。
【Abstract】 Objective To investigate the protective mechanism of Angelica sinensis polysaccharide on liver cell injury in mice. Methods Acute liver injury model was established by one-time irrigation stomac. The mice were randomly divided into angelica polysaccharide group, alcohol model group and normal control group at the same time. Angelica sinensis polysaccharide group was injected Angelica Polysaccharide on continuous 6 days; The alcohol model group and normal control group were injected with the same amount of normal saline. Observed the general situation and liver tissue morphological structure changes. Malondialdehyde, superoxide dismutase and glutathione peroxidase were detected in serum of all groups. Results Angelica Polysaccharide group was recovered significantly more than alcohol model group; Angelica Polysaccharide group was repaired completely about 11 days, There was no change in the structure of liver tissues in normal control group. Serum biochemical test showed that malondialdehyde, superoxide dismutase and glutathione peroxidase were statistically significant between angelica polysaccharide group and alcohol model group. Conclusion Angelica sinensis polysaccharides can improve antioxidant enzyme activity. It also can remove oxygen free radicals and reduce membrane lipid peroxidation chain reaction. Therefore, the structure of liver cell membrane was restored. Angelica sinensis polysaccharide could protect liver cells.
